摘    要:目的研究活血通络汤对激素性股骨头缺血性坏死(SANFH)家兔血浆内皮素(ET)、骨钙素(BGP)的影响。方法将96只13本大耳兔随机分为正常组(24只)和造模组(72只),按贺氏造模法造模后各随机抽取8只,两组对比检测造模是否成功。成功后再将造模组随机分为活血通络汤高、中、低剂量治疗组及模型组,治疗组按高、中、低剂量分别灌服活血通络汤,模型组和正常组均灌服等量蒸馏水。于给药第8W末,进行血浆ET、BGP的检测,同时电镜观察股骨头组织空骨陷窝数。结果与正常组比较,模型组血浆ET含量升高(P〈0.05),血浆BGP含量降低(P〈0.05),股骨头空骨陷窝率升高(P〈0.05);与模型组比较,高、中、低剂量组血浆ET含量明显降低(P〈0.01),血浆BGP含量明显升高(P〈0.01),股骨头空骨陷窝率明显降低(P〈0.01);高、中、低剂量组间比较,剂量越高疗效越优(P〈0.05)。结论活血通络汤能够明显下调SANFH模型兔血浆ET含量,提高血浆BGP含量,降低空骨陷窝率,提示活血通络汤在改善缺血环境的同时,可促进坏死骨的修复和新骨的形成。

Abstract:Objective To study the effects of Huoxue Tongluo Decoction on the plasma endothelin (ET) and protein (BGP) of rabbits with steroid-induced avascular necrosis of the femoral head ( SANFH ). Methods Ninety six Japanese white rabbits were randomly divided into normal group (24 ones) and model group (72 ones). After the establishment of model by the Hayes method, 8 rabbits were randomly selected from each group. Comparison was made whether the model was successfully established in the two groups. After the successful modeling ,the rabbits in the model group were randomly divided into treatment groups with high, medium, and low dose of Huoxue Tongluo Decoction and model group. The treatment groups received the drench with high, medium, and low dose of Huoxue Tongluo Decoction,respectively. While the model and normal groups received the drench with the same volume of distilled water. At the end of the eighth week, plasma ET and BGP were detected, and the number of empty lacuna in femoral head tissues was observed by electron microscope. Results The content of plasma ET in the model group significantly increased ( P 〈 0.05 ), and its content of plasma BGP and the number of empty lacuna significantly increased (P 〈 0.01 ) compared with those in the normal group. Compared with the the model group, the content of plasma ET decreased ( P 〈 0.05 ) , and the content of BGP significantly increased ( P 〈 0.01 ) in the high, medium, and low dose groups. The number of empty lacuna also decreased significantly ( P 〈 0.01 ). Among the high, medium, and low dose groups, the higher the dose, the superior the curative effect ( P 〈 0.05 ). Conclusion Huoxue Tongluo Decoction can obviously down-regulate the plasma ET content in SANFH rabbit models, increase the content of plasma BGP, and reduce the rate of empty lacunae, which indicates that Huoxue Tongluo Decoction can not only improve the ischemic environment, but also promote the repair of the necrotic bone and the formation of new bone.
