当归注射液对免疫诱导再生障碍性贫血小鼠骨髓细胞CD_(34)~+及Fas抗原表达的影响

摘    要:目的探讨当归注射液对免疫诱导再生障碍性贫血小鼠骨髓细胞CD34+及Fas抗原表达的影响。方法建立免疫诱导再障小鼠模型,随机分为当归组、再障组、正常组,分别测定各组小鼠的体重、外周血白细胞计数、骨髓单个核细胞计数以及流式细胞仪检测骨髓单个核细胞CD34+及Fas抗原表达的荧光强度,应用SPSS10.0统计软件进行分析。结果再障组及当归组小鼠的体重、外周血白细胞计数、骨髓单个核细胞计数较正常组均有降低,但当归组明显高于再障组(P<0.05),当归组骨髓细胞CD34+抗原表达的荧光强度(69.0±8.3)明显高于再障组(33.6±9.1,P<0.05),而与正常组(78.2±9.4)之间无显著性差异(P>0.05)。再障组骨髓单个核细胞Fas抗原表达的荧光强度(62.7±7.1)明显高于正常组(21.5±3.6,P<0.05),当归组骨髓单个核细胞Fas抗原表达的荧光强度(43.0±6.8)明显低于再障组(62.7±7.1,P<0.05)。结论当归能降低再障小鼠骨髓细胞Fas抗原的表达,减少细胞凋亡,促进骨髓造血细胞增生,有刺激造血作用。

Abstract:Objective To investigate the effect of Angelicae Sinensis injection on CD34+ and Fas antigen expression of bone marrow cells in immune-induced aplastic anemia (AA) Mice. Methods The model of immune-induced AA mice were induced by means of 6.0 Gy 60Co γ-ray irradiation and lymphocyte infusion through tail vein. The mice were divided into 3 groups,the Angelicae Sinensis group,the AA group and the normal group. To measure the weight,the count of blood leucocyte and bone marrow nucleated cell,CD34+ antigen expression of bone marrow cells,Fas antigen expression of bone marrow cells. All dam analyzed by SPSS 10.0. Results CD34+ antigen expression intensity of Radix Angelicae Sinensis group was 69.0±8.3 and much higher than the AA group's(33.6±9.1,P<0.05),with no statistic difference from that in normal group (80.5±2.6,P>0.05). Fas antigen expression intensity of the AA group was 62.7±7.1 and much higher than the normal group's(21.5±3.6,P<0.05). Fas antigen expression intensity of Angelicae Sinensis group was 43.0±6.8 and much lower than the AA group's(62.7±7.1,P<0.05). Conclusion Angelicae Sinensis could promote proliferation of stem and progenitor cell of AA mice.
