小鼠螺旋神经节神经元的分离和A型钾通道的电生理特性

摘    要:
目的机械分离并酶解小鼠耳蜗螺旋神经节神经元(SGN)细胞,观察其A型钾通道的电生理特性。方法对生后1~6d的小鼠耳蜗螺旋神经节组织进行机械分离并酶解,对SGN进行免疫细胞化学染色鉴定,并利用全细胞膜片钳技术记录和分析A型钾通道电流。结果新生小鼠SGN在机械分离和酶解条件下,可以获得良好的细胞形态。SGN的A型钾通道在-60mV时激活,+40mV时失活,能被细胞外的4-氨基吡啶阻滞。结论成功建立了机械分离并酶解SGN细胞的方法,SGN的A型钾通道具有激活快、失活快的特点。
