摘    要:
目的:探讨促红细胞生成素(Erythropoietin,EPO)对新生大鼠缺氧缺血性脑损伤(hypoxic-ischemic brain damage,HIBD)的保护作用及其可能的机制。方法:新生7天大鼠随机分3组:假手术组、HIBD模型组和EPO治疗组(HIBD+EPO组)。观察缺氧缺血后脑组织含水量、病理改变、免疫组织化学检测凋亡因子Bcl-2的表达以及脑萎缩程度。结果:EPO治疗能显著减轻缺氧缺血侧(左侧)大脑半球水肿、病理学改变和大脑半球萎缩程度。左大脑半球含水量降至(85.93±1.19)%,显著低于HIBD模型组(86.75±0.87)%(P<0.05),左脑组织萎缩比(14.95±14.15)%,显著低于模型组(28.26±19.39)%(P<0.01)。HIBD+EPO组大鼠缺血侧皮层Bcl-2阳性细胞表达较HIBD模型组及假手术组均显著增多。结论:EPO对新生鼠缺氧缺血性脑损伤具有保护作用,其机制与提高抑制细胞凋亡的发生有关。

Neuroprotective effect of erythropoietin on hypoxic-ischemic brain damage in neonatal rats
Abstract:
Objective:To study the neuroprotective effect of erythropoietin on hypoxic-ischemic brain injury in neonatal rats.Methods:7 day old rats were randomly divided into 3 groups:sham operation group,HIBD model group and erythropoietin group.Water content of the brain,changes of brain histopathology,bcl-2 protein expression and extent of brain atrophy were observed.Results:Erythropoietin treatment significantly mitigated brain edema,pathological lesions and the extent of atrophy of hypoxic-ischemic hemisphere.The water content of left hemisphere was reduced to(85.93±1.19)%,significantly lower than that of HIBD model group(86.75± 0.87)%(P<0.01).The atrophy ratio of left hemisphere in erythropoietin group was(14.95±14.15)%,significantly lower than that of HIBD model group(28.26±19.39)%(P<0.01).And there was obvious diference in the expression of bcl-2 protein in cortex between EPO group and HIBD model group and sham operation group(P<0.01).Conclusion:Erythropoietin is neuroprotective against hypoxic-ischemic brain injury in neonatal rats,and up-regulating bcl-2 expression might contribute to the protective mechanism.
