硫化氢对腹主动脉缩窄致心肌损伤大鼠心肌组织COXII表达的影响

摘    要:目的:探讨细胞色素C氧化酶II(COXII)在腹主动脉缩窄所致的心力衰竭组织中的表达变化及硫化氢(H2S)干预的影响。方法:30只健康雄性SPF级SD大鼠随机分为对照组、心衰组和H2S干预组,每组10只。通过腹主动脉结扎术制作大鼠心衰模型,术后12周,采用动物超声监测舒张末室间隔厚度(IVS)、舒张末期左心室后壁厚度(LVPW)、左心室射血分数(LVEF);血流动力学监测左心室收缩压力(LVSP)、左室舒张末压(LVEDP)、左室内压上升或下降最大速率(LV±dp/dtmax);记录左心室质量指数(LVMI);苏木精-伊红及Masson染色观察心肌组织病理变化;采用Real-time PCR及Western blot检测心肌组织中COXII的表达。结果:与心衰组比较,H2S干预组大鼠LVMI、IVS、LVPW、LVEDP及LVEF明显降低,而LVSP、LV±dp/dtmax显著升高(P〈0.05)。苏木精-伊红及Masson染色显示心肌损伤程度减轻。Real-time PCR及Western blot结果显示干预组大鼠心肌组织中COXII mRNA及蛋白明显高于心衰组(P〈0.05)。结论:心肌组织中COXII表达升高在心肌衰竭的发病机制中起到重要作用,而H2S可以改善心力衰竭的心肌损伤程度,其机制可能与提高COXII蛋白的表达有关。

The effect of hydrogen sulfide on the expression of COXII in myocardial injury induced by coarctation of the abdominal aorta in rats

Abstract:Objective:To study the expression of COXII in heart failure induced by coarcation of the abdominal aorta in rats and to investigate the effect of hydrogen sulfide on heart failure.Method:30 male SD rats were randomly divided into three groups including control group,heart failure group and hydrogen sulfide intervention group.The abdominal aorta in rats were ligated to establish the heart failure model.12 weeks later Hemodynamic Parameters including LVSP,LVEDP and LV±dp/dtmax,animal ultrasonography results and histopathological changes including HE staining and Masson staining were observed in all rats.The expression of COXII protein were detected using Real-time PCR and Western blot methods.Result:Compared with the heart failure group,the thickness of left ventricle in H_2S group was significantly decreased.Hemodynamic Parameters such as IVS,LVPW,LVEF and LV±dp/dtmax were significantly improved(P0.05).The grade of histopathological damage of heart tissue were attenuated greatly.The expression of COXII mRNA and protein were significantly higher in H_2S group than heart failure group(P〈0.05).Conclusion:Increasing of COXII expression in heart tissue might play an important role in the heart failure.Hydrogen sulfide could attenuate the degree of heart failure in rats.The mechanism might be associated with the up-regulation of COXII expression.
