无肝期大鼠小肠异丙酚代谢相关UGT1A6的基因表达变化

摘    要:目的研究无肝期前后与异丙酚代谢相关的UGT1A6在大鼠小肠基因表达的变化,以期初步阐释异丙酚无肝期肝外代谢特点形成的原因.方法 15只大鼠随机分为3组(5只/组),A组为对照组,B组阻断肝门30min,C组阻断肝门60min,截取各组大鼠小肠组织,采用逆转录聚合酶链式反应(RT-PCR)技术,检测各组UGT1A6mRNA的表达.结果大鼠小肠组织UGT1A6的基因表达水平B、C组明显高于A组(P<0.01);B组与C组相比,没有显著差异(P>0.05).结论大鼠小肠组织UGT1A6基因表达在无肝期比无肝期前增多,这可能是异丙酚无肝期肝外代谢增强的机制.

Abstract:Objective Gene expression variation of small intestine UGT1A6 of rat related propofol metabolism was investigated in anhepatic phase to primarily explain the reason of extrahepatic metabolism characters of propofol.Methods Fifteen rats were distributed to 3 groups(5/group) randomly, control group was named A, 30min post-devascularization of the hepatic portal was B group, 60min post-devascularization of the hepatic portal was C group. Small intestine tissues of rat were taken and UGT1A6mRNA were detected by RT-PCR.Results Gene expression levels of small intestine UGT1A6 of B and C group were significantly higher than A group's(P<0.01);The gene expression levels between B and C were no significantly differences(P>0.05).Conclusion Gene expression levels of small intestine UGT1A6 of rat are increased in anhepatic phase than in hepatic portal dissociation phase, which is the possible reason that extrahepatic metabolism of propofol is increased in anhepatic phase.
