神经生长因子与人胆管癌神经浸润转移的相关性分析

摘    要:目的探讨神经生长因子(nerve growth factor,NGF)及其酪氨酸激酶受体A(TrkA)与人胆管癌神经浸润、淋巴转移和病理类型的关系。方法采用免疫组织化学技术,检测92例人胆管癌组织中β-NGF和TrkA的表达情况,Spearman’s等级相关检验β-NGF的表达与胆管癌神经浸润、淋巴转移以及病理分型的关系。结果β-NGF和TrkA均在胆管癌组织中高表达,二者存在相关性,且在神经浸润组和无神经浸润组的表达差异有统计学意义;NGF阳性表达随着肿瘤恶性程度的增高而增高,且与肿瘤的淋巴转移密切相关。TrkA还表达于神经鞘膜。结论NGF与胆管癌神经浸润密切相关。NGF可能通过旁分泌途径与TrkA作用促使癌细胞沿神经转移;通过自分泌作用促进胆管癌细胞增殖。

Abstract:Objective To investigate the role of nerve growth factor (NGF) and its receptor TrkA in the perineural invasion,lymph node metastasis and pathological types of human cholangiocarcinoma.Methods The expression of NGF and TrkA was detected by SP immunohistochemical technique in 92 cases of cholangiocarcinoma.Spearman's correlation test was used to analyze the relationship between the NGF positive expression and perineural invasion,lymph node metastasis as well as pathological types.Results Both NGF and TrkA were highly expressed in cholangiocarcinoma tissues.The positive expression rate of NGF was much higher in perineural invasion group than in non-perineural invasion group,and the difference was significant.Furthermore,the NGF positive expression was also strongly associated with lymph node metastasis and poor differentiation.TrkA was also positively expressed in perineurium.Conclusion NGF may enhance the proliferation of cholangiocarcinoma cells by autocrine loop,and stimulate the perineural invasion by paracrine loop.
