c-myc、MMP-9在受压静脉管壁的表达

摘    要:
目的:研究c-myc蛋白和MMP-9在受压静脉管壁的表达,初步探讨静脉受压后血管重塑的分子机制.方法:42只SD大鼠随机分为对照组(21只)和实验组(每时间点各3只,共21只),用5#注射针头辅助作部分髂静脉结扎造模.分别在0.5、12、24、48、72h、7和14d取材,明胶液包埋后冰冻切片,用免疫组化SP法检测c-myc蛋白和MMP-9的表达,采用计算机图像分析系统测定切片的平均积分光密度(IOD).结果:对照组和正常静脉管壁均未见c-myc蛋白、MMP-9表达,实验组c-myc蛋白从12 h开始表达,MMP-9从24 h开始表达上调,并逐渐增强,IOD逐渐升高,与对照组比较,差异具有显著性(P<0.05).7 d时两者表达同时达到高峰.c-myc蛋白与MMP-9在受压静脉管壁的表达呈正相关(r=0.7532,P<0.05)结论:c-myc基因产物和MMP-9在受压静脉管壁表达增强,两者在受压静脉管壁重塑过程起着重要作用;c-myc基因和MMP-9的表达具有相关性.

The expression of c-myc and MMP-9 in compresed common iliac vein of rats

Abstract:
Objective: To explore the expression change of c-myc and MMP-9 in compressed common iliac vein of rats for exploring molecular mechanism of vascular remodeling in the compressed vein. Methods: 42 adult SD rats were divided into 2 groups randomly: control group (21 rats) and experimental group (21 rats,3 rats for different detection times). The model of vein iigation was established by assisting of 5# of needle. The compressed segments of common iliac vein,including the proximal and distal part of the compressed vein were taken at 0.5h,12h,24h,48h,72h,7d,and 14d after operation. Immunohistochemistry was used to detect the expression ofc-myc and MMP-9,while expression level was analyzed by the integral optical density (IOD) of slices adopting biomedical image analysis system,Results: There was no positive expression of the c-myc and MMP-9 in the vessel wall of control group. The expression of c-myc in the vessel wall of experimental group appeared at 12h after compression,and reached its peak at the 7th day after compression. However,the expression of MMP-9 appeared at 24h after compression,and reached its peak at the 7th day after compression. The IOD increased gradually with the duration of compression time,comparing with that of the control (P<0.05). The expression of c-myc was positively correlated with that of MMP-9 (r=0.7532,P<0.05). Conclusions: c-myc and MMP-9 perhaps play an important role in vascular remodeling of compressed vein.
