安心颗粒对心力衰竭大鼠细胞凋亡线粒体超微结构的影响

摘    要:
目的:观察安心颗粒对阿霉素诱导的慢性心力衰竭大鼠心肌细胞线粒体超微结构的影响,探讨其可能的机制。方法:将50只雌性SD大鼠随机分为正常对照组(A组)、模型组(B组)、卡托普利组(C组)、安心颗粒小剂量组(D组)、安心颗粒大剂量组(E组),每组10只。B,C,D,E组给予阿霉素腹腔注射造模,造模同时C,D,E组灌胃给药。连续给药6周后,测定大鼠血清SOD和MDA含量;用原位末端标记法检测大鼠心肌细胞凋亡,并计算凋亡指数。在电子显微镜下观察大鼠心肌组织切片线粒体超微结构。结果:安心颗粒大剂量组可以升高血清SOD水平,降低MDA水平,增加心肌细胞线粒体的数量及减少心肌细胞线粒体损害,抑制细胞凋亡。结论:安心颗粒对心衰大鼠线粒体的损害有一定的保护作用,其作用机制可能与减轻氧化应激损伤、保护心肌细胞线粒体结构与功能、抑制心肌细胞凋亡等有关。

Abstract:
Objective: To observe the effects of Anxin granules on apoptosis mitochondria ultratructure in cadiocyte of rats with chronic heart failure which was induced by adriamycin and discuss the possible of mechanism. Methods: Fifty female SD rats were randomized into normal control group(Group A), model group(Group B), captopril group(Group C), low dosage Anxin granules group(Group D), high dosage Anxin granules group(Group E), in which each has ten rats. B, C, D and E group were given adriamycin peritoneal injection to establish the model, C, D and E group were given gastric infusion at the same time. After six weeks successive administration, SOD and MDA contents in rats' serum were determined; myocardial apoptosis of rats were observed using an in situ end labeling method and apoptosis index was calculated. Mitochondria ultratructure of myocardium tissue sections in rats was observed in electron microscope. Results: The high dosage Anxin granules group could increase the level of SOD, decrease the level of MOD, also could increase the quantity and decrease the injury of myocardial cells mitochondria, inhibit apoptosis. Conclusions: Anxin granules has certain protective effects on mitochondria injury in rats with heart failure, the mechanism probably related to reduce oxide stress injury, protect structures and functions of myocardial cells mitochondria, inhibit apoptosis of myocardial cells, etc.
