丁苯酞对脑缺血再灌注模型大鼠海马组织中超氧化物歧化酶和特异性半胱氨酸蛋白酶-3表达的影响

摘    要:
目的:研究丁苯酞对脑缺血再灌注模型大鼠海马组织中超氧化物歧化酶(SOD)和特异性半胱氨酸蛋白酶-3(Caspase-3)表达的影响。方法:取大鼠用四血管闭塞法建立脑缺血再灌注模型,选取神经功能缺损评分1~3分的大鼠,随机分为模型组(食用麻油)、阳性对照组(尼莫地平10mg·kg-1)、治疗组(丁苯酞25mg·kg-1),另设立假手术组(食用麻油);每组12只,灌胃给予相应药物,每天1次,2周后行神经功能缺损评分,断头取脑,检测海马组织中SOD与Caspase-3活性变化情况。结果:与模型组比较,阳性对照组和治疗组大鼠神经功能缺损评分2周后明显降低,海马组织中SOD活性明显升高,Caspase-3表达明显降低(P均<0.05),其中阳性对照组与治疗组之间比较无显著性差异(P>0.05)。结论:丁苯酞可通过上调模型大鼠海马组织中SOD活性和下调Caspase-3表达发挥对脑缺血再灌注脑损伤的保护作用。

Abstract:
OBJECTIVE: To study the effect of butylphthalide (NBP) on the expression of SOD and Caspase-3 in hippocampus tissue of rats with cerebral ischemia-reperfusion. METHODS: Four-vessel occlusion was used to build cerebral ischemia-reperfusion models. Model rats with neurologic impairment scores of 1~3 grade were randomly divided into model group (edible castor oil), positive control group (nimodipine10 mg·kg-1), treatment group (NBP 25 mg·kg-1) with each group of 12 rats. Sham operation group was also set up (edible castor oil). Each group was given relevant drugs once a day. Two weeks later, neurologic impairment scores were recorded and the expression of SOD and Caspase-3 in hippocampus tissue were detected. RESULTS: Compared with model group, the neurologic impairment scores of positive control group and treatment group decreased significantly, the activity of SOD hippocampus tissue increased while the expression of Caspase-3 decreased (P0.05). There was no significant difference between positive control group and treatment group (P0.05). CONCLUSION: NBP can significantly up-regulate the activity of SOD and down-regulate the expression of Caspase-3 to protect tissue from the ischemia injury.
