HPLC-ELSD法测定柏子仁霜中胡萝卜苷的含量

目的:建立测定柏子仁霜中胡萝卜苷含量的方法,并比较不同产地柏子仁炮制后的霜品中胡萝卜苷的含量。方法:采用高效液相色谱-蒸发光散射检测法。色谱柱为Scienhomeinertex C18(150mm×4.6mm,5μm),流动相为甲醇-水(80∶20),流速为1.0mL·min-1,柱温为30℃;载气为氮气,气体压力为30.0psi,漂移管温度为80℃。结果:胡萝卜苷进样量在0.77～4.45μg范围内,其对数值与峰面积对数值呈良好线性关系(r=0.9997);平均回收率为96.55%,RSD=1.68%(n=6)。不同产地柏子仁霜中胡萝卜苷含量分别为0.4070%(河南辉县)、0.3281%(安徽淮北)、0.3384%(山东枣庄)、0.3261%(山东长清)。结论:本方法稳定、简便、快速,适用于柏子仁霜的质量控制。

Content Determination of Daucosterol in Platycladus orientalis Cream by HPLC-ELSD

OBJECTIVE：To establish the method for the content determination of daucosterol in Platycladus orientalis cream,and to compare the content of daucosterol in the cream of P.orientalis from different producing area after processed.METHODS：HPLC-ELSD method was adopted.Scienhome inertex C18（150 mm×4.6 mm,5 μm）was used with methanol-water（80：20） at flow rate of 1.0 mL·min-1.The column temperature was set at 30 ℃ and nitrogen was as carrier.The pressure of gas was 30.0 psi and the temperature of drift tube was 80 ℃.RESULTS：The linear range of daucosterol was 0.77-4.45 μg（r=0.999 7） with an average recovery of 96.55%（RSD=1.68%,n=6）.The contents of daucosterol in P.orientalis cream were 0.407 0%（from Huixian county of Henan province）,0.328 1%（from Huaibei of Anhui province）,0.338 4%（Zaozhuang of Shandong province）,0.326 1%（Changqing of Shandong province）,respectively.CONCLUSION：This method is stable,simple and rapid for the quality control of P.orientalis cream.
