大鼠急性胰腺炎中核因子NF-κB抗凋亡机制的研究

摘    要:
目的探讨核因子NF—kB激活在急性胰腺炎抗凋亡机制中的作用。方法利用原位细胞凋亡检测法和免疫组织化学方法检测大鼠胰腺细胞凋亡及Bcl-2和Bcl—XL在胰腺组织中的表达。结果预防或治疗性给予N-乙酰半胱氨酸可不同程度地降低NF—kB激活,减轻胰腺坏死程度,增加胰腺腺泡细胞凋亡程度,降低Bcl-2和Bcl—XL蛋白的表达。结论细胞凋亡对腺泡细胞具有保护作用,与急性胰腺炎的严重程度呈负相关。Bcl-2和Bcl—XL基因表达可能参与NF—kB抗胰腺腺泡细胞凋亡过程。

Abstract:
[Objective] To investigate the anti-apoptosis mechanism of nuclear factor NF-kB activation in experimental acute pancreatitis. [ Methods] Appliation of TUNEL for detecting apoptosis in rat pancreatic acinar cells, and SABC immunohistochemical technique for detecting the expression of Bcl-2 and Bcl-XL genes. [Results] The pre- and post-treatment with NAC can remarkably depress the activation of NF-kB,relieve the degree of pancreas necrosis ,improved the degree of acinar cells apoptosis and lower the expression of Bcl-2 and Bcl-XL. [Conclusions] Apoptosis can protecet the pancreas cell and the degree of apoptosis is inversely related to the severity of acute pancreatitis. The expression of Bcl-2 and Bcl-XL genes might take part in the anti-apoptosis mechanism of NF-kB activation in rats with experimental acute pancreatitis.
