SIRT1在乳腺癌中的作用

沉默信息调节因子1(sirtuin Type 1,SIRT1)是酵母Sir2的哺乳动物同源体,是一种NAD+依赖的Ⅲ类组蛋白去乙酰化酶,能使组蛋白和非组蛋白去乙酰化,参与细胞多种生理、病理过程,如基因沉默、细胞衰老、寿命延长、抗氧化应激、能量代谢调节、DNA损伤修复、肿瘤发生等.其中SIRT1在乳腺癌的发生中涉及的上下游信号分子众多,信号通路繁杂而交叉.有研究显示SIRT1起着抑制乳腺癌发生的作用;而有的研究结果正相反,SIRT1起着促进乳腺癌发生的作用.

Roles of SIRT1 in breast cancer

Sirtuin Type 1(IRT1) is a mammalian homolog of yeast Sir2.It is a nicotinamide adenine dinucleotide(NAD+)-dependent classⅢ histone deacetylase which removes acetyl groups from histones and many non-histone proteins.Therefore it is involved in various physiological and pathological processes in cells such as gene silencing,cell aging,life extension,stress response,energy metabolism,DNA damage repairing,and tumorigenesis.In recent years,many studies have shown that SIRT1 is involved in the occurrence and development of breast cancer.SIRT1 interacts with many upstream and downstream signaling molecules,participating in many complex signaling pathways and cross-pathways.Some studies have shown that SIRTl plays a role in inhibiting breast cancer;on the contrary,the others have shown that SIRTl plays a role in promoting breast cancer.
