摘    要:
系统性红斑狼疮(systemic lupus erythematosus,SLE)是一种典型的系统性自身免疫性疾病,T淋巴细胞在机体大量自身抗原刺激下,信号传递及基因表达出现多种特异性缺陷,在疾病发生发展中起到关键作用。CD3复合物的重构及胞膜表面脂筏的大量聚集使SLET细胞的活化阈降低,胞内信号级联紊乱;同时,聚集的脂筏中黏附分子表达上调促使T细胞向靶组织定向趋化;信号转导紊乱导致转录因子表达改变,DNA甲基化异常引发基因表达失衡。由此,功能异常的T细胞中改变的多种信号分子可以作为诊断和治疗SLE的潜在性分子靶标。

Abstract:
Systemic lupus erythematosus(SLE) is a classic systemic autoimmune disease.T cells from patients SLE exhibit several discrete and specific defects that alter signaling pathways,the gene expression pattern and behavior upon stimulation.Rewiring of the CD3 complex and aggregation of surface-membrane lipid rafts grant SLE T cells a lower activation threshold and distort the ensuing signaling events.Additionally,increased expression of adhesion molecules within aggregated lipid rafts guides them to target organ...
