超声微气泡介导cy5ODN转染大鼠肾的研究

摘    要:目的:探讨超声联合脂质体微气泡介导寡核苷酸cy5ODN靶向转染肾的有效性和安全性。方法:健康雄性Wistar大鼠30只,分为对照组、单纯cy5ODN组、造影剂+cy5ODN组、超声+cy5ODN组和超声+造影剂+cy5ODN组。予以实验刺激后,7d取肾组织检测荧光表达,光镜下观察肾组织病理改变,以评价该方法的副作用。结果:超声+造影剂+cy5ODN组cy5ODN基因转染率显著高于其余各组,实验各组均未见到明显肾小球损害。结论:超声联合类脂质体微泡能显著增强基因在肾组织的转染效率,可望为肾疾病的基因治疗提供一种新的安全、有效的转导方法。

Abstract:Objective To study the efficiency and safety of transduction of oligodeoxynuleotides eySODN to kidney by ultrasounds mediated liposome microbubbles. Methods Thirty healthy rats were divided into 5 groups., control group, simple cySODN group, contrast agent plus cySODN group, ultrasound plus cySODN group, ultrasound plus contrast agent plus cySODN group. Laser focal concentration microscope was used to detect expression of fluorescence, and the pathological changes of kidney were observed with light microscope to evaluate the side effects of this method. Results Transefection efficiency in ultrasound plus contrast agent plus cySODN group was significantly higher than other groups and no obvious damage of glomcrulus was observed in all groups. Conclusions As a new type of gene introduction measure,ultrasound-activated microbubbles could significantly improve the transefection efficiency of gene and it is safe and easy to use with good targeting ability and should be used widely.
