Monomeric circular λ DNA molecules were isolated from phage-infected cells, purified, and their biological activity tested using in vitro DNA packaging and λ terminase assays. These circles, which contain a single cos, proved to be as efficient as linear λ concatemers both for packaging into viable phage and cleavage at cos by highly purified λ terminase. |
