小鼠静脉,腹腔用药后顺铂结合瘤细胞DNA的定量比较研究

腹水瘤小鼠腹腔或静脉顺铂给药后，提取腹水瘤细胞的ＤＮＡ，应用石墨炉原子吸收光谱法测定与ＤＮＡ结合的总铂量。结果发现；腹腔给药后４小时，２４小时及４８小时ＤＮＡ的总铂量高于静脉给药。

A Comparison of quantitation of Platinum-DNA Binding following the Iv and Ip Routes of administration of cisplatin in ascitic hepatoma mice

Cisplatin at a dose of 35mg/kg was administered to mice ascitic hepatoma either Iv or Ip. The DNA was isolated from hepatoma cell pellets of each of the Balb/c mice.The amount of total platinum binding to DNA was measured by atomic absorption spectroscopy(AAS). The data showed:platinum binding to DNA levels following Iv administraton were lower than that of follwing Ip at 4 hours,24 hours and 48 hours. There was a significant difference in total platinum binding DNA between following Ip administration and folliwing Iv administration at 24 hours and 48 hours.And total platinum-DNA binding was remarkably reduced(P<0.05)with time going(from 4 hours to 48 hours) in followingIv administration.There was a slower reduction in total platinum DNA(P>0.05)in following Ip administration.The result indicated that Ip chemotherapy enhanced cisplatinum induced cytotoxicity in mice ascitic hepatoma cells.
