促红素对慢性肾功能不全脂质紊乱疗效的实验研究

摘    要:目的 证实重组人促红素(rhEpo)对慢性肾衰竭脂质紊乱的治疗作用,并观察其对慢性肾衰竭鼠的病理学影响。方法 应用5/6肾切除法制成慢性肾功能不全(CRF)大鼠模型,术后给予rhEpo,用药后测定其内生肌酐清除率、尿蛋白量、血清脂质和红细胞比积,并取肾组织做病理检查。结果 与正常对照组相比,行5/6肾切除术的大鼠血肌酐浓度明显升高(P<0.01),内生肌酐清除率明显降低(P<0.01);模型对照组甘油三酪、胆固醇及低密度脂蛋白明显增高(P<0.01)。与模型对照组相比,模型治疗组的红细胞比积明显升高(P<0.01),血清脂质中,甘油三酪、胆固醇和低密度脂蛋白的浓度明显降低(P<0.01)。病理学观察,模型对照组和模型治疗组之间未见明显差别。结论 重组人促红素能显著缓解CRF大鼠模型的脂质紊乱,而对CRF鼠模型的病理改变无显著作用。

Abstract:Objective To testify the therapeutic effects of the recombinant human erythropoietin (rhEpo) on the lipid disorders of chronic renal failure (CRF).To observe the possible function of rhEpo on the pathology of CRF.Methods Twenty four male Wistar rats were subjected to 5/6 nephrectomy(CRF group and Epo CRF group) or sham operation(NL group).The Epo CRF group was given rhEpo twice a week on a dose of 150IU/kg. Then, the subgroups of animals were studied. Results Compared with the NL group, concentrations of TG,CHO and LDL were markedly increased in the CRF group( P <0.01 respectively).At the same time, in contrast to the CRF group, these concentrations of the Epo CRF group had significant improvements( P <0.01 respectively).There was no significant pathological distinction between the CRF group and Epo CRF group. Conclusion Besides correcting the Epo deficiency anemia,the therapy of rhEpo also has positive effects on the serum lipid disorders of CRF rat models, but it seems to have no obvious influence on the pathology of CRF.
