摘    要:目的:研究三氧化二砷(arsenic trioxide,As2O3)对体外培养的人类风湿性关节炎(rheumatoid arthritis,RA)滑膜细胞凋亡诱导作用。方法:酶消化法进行人关节滑膜细胞原代培养,MTT法研究As2O3对RA滑膜细胞活力影响;用光镜、电镜、DNA凝胶电泳等方法检测As2O3诱导RA滑膜细胞凋亡。结果:成功体外培养人关节滑膜细胞。MTF法显示As2O3可抑制RA滑膜细胞活力;光镜、电镜下发现As2O3作用于RA滑膜细胞可出现凋亡表现;DNA凝胶电泳显示有凋亡梯带出现。结论:As2O3可以诱导RA滑膜细胞凋亡。

Abstract:Objective: To investigate the effects of arsenic trioxide (As2O3) on apoptosis of synoviocytes in human rheumatoid arthritis (RA).Methods: Primary synovocytes were cultured by enzymatic digestion. The inhibition of synoviocytes viability was detected by MTT method. Apoptosis changes induced by As2O3 were examined by light microscope, electron microscope and DNA gel electrophoresis. Results: Synoviocytes were cultured successfully in vitro. The viability of synoviocytes could be inhibited by As2O3 which was confirmed by MTT method. Apoptosis was detected by light microscope and electron microscope. Apoptosis Ladder was seen by DNA gel electrophoresis. Conclusion: As2O3 can induce apoptosis of synoviocytes in RA.
