摘    要:目的探讨磺酰罗丹明B法检测多巴胺能神经细胞的最佳条件。方法采用系列细胞密度梯度、选用多个波长、加用参考波长,以磺酰罗丹明B法检测多巴胺能细胞系Mes23.5细胞数量和活性,分析细胞数量与检测波长及吸光度的关系。结果细胞数量与吸光度呈线性关系,最佳检测波长为540nm。结论磺酰罗丹明B法检测多巴胺能细胞结果可靠,是切实可行的该细胞定量分析和活性检测方法。

Abstract:Objective To explore the optimal detecting conditions of the dopaminergic neural cell by SRB assay. Methods A series of cell density grads, sereval working wavelengths, and reference wavelengths were chosen to detect the number and the activity of the dopaminergic neural cell Mes23. 5 by SRB assay. The relations between cell number and the absorbency were analysed. Results The relations between cell number and the absorb-ency were linear, and the optimal working wavelength was 540nm. . Conclusion The detecting result of the dopaminergic neural cell by SRB assay is reliable, so SRB assay is a practical method to quantitatively analyse the dopaminergic neural cell and detect their activity.
