IL-28 B基因多态性与新疆维吾尔族HCV易感性研究

摘    要:目的:探讨白介素-28B( IL-28B)基因的多态性与新疆维吾尔族慢性丙型肝炎感染易感性的关联。方法选取180例新疆维吾尔族慢性丙肝患者作为HCV组,150例维族健康体检者作为健康对照组,通过PCR产物测序法检测两组受试者IL-28B rs8099917、rs12979860基因型和等位基因分布频率,对两组基因型和等位基因差异进行分析;同时在HCV组中,对不同IL-28B基因型人群的HCV-RNA载量、肝功能、血脂等进行分析。结果①rs8099917在HCV组和健康对照组中TT和TG/GG基因型频率差异有统计学意义(χ2=11.627,P=0.001,OR=1.363,95%CI:1.107~1.591);G等位基因率在两组间差异明显,HCV组中占10.6%,健康对照组0.17%(χ2=7.011, P =0.008, OR =0.658,95%CI:0.512~0.905);② rs12979860在两组间的基因型和等位基因频率差异无统计学意义;③rs12979860 TT型患者的HCV-RNA高于TG/GG型,差异有统计学意义( P=0.032);患者肝功血脂差异与IL-28B基因多态性中无关。结论新疆维吾尔族人群中HCV的易感性与IL-28B rs8099917基因多态性有关联,rs8099917中等位基因G是新疆维族HCV患者的易感基因。

Genetic polymorphisms of interleukin-28 B associated with susceptibility to hepatitis C virus infection
Abstract:Objective To study the association between interleukin-28B (IL-28B) single nucleotide polymorphism and susceptibility to hepatitis C virus ( HCV) infection in Xinjiang Uyghur populition. Methods In this study,the IL-28 B rs8099917 and rs12979860 locus were genotyped in 180 patients infected with HCV and 150 healthy con-trols using the method of direct gene sequencing. To analyze the differences of SNPs genotypes and allele frequen-cies in two groups,SPSS 13. 0 was used. Results ① TT and TG/GG genotype frequencies of rs8099917 differed significantly between patients with chronic HCV and healthy controls (χ2 =11. 627,P=0. 001,OR=1. 363,95%CI:1. 107 ~1. 591 ) . Patients with HCV infection had a higher G allele frequency than healthy controls (χ2 =7. 011,P=0. 008,OR=0. 658,95%CI:0. 512~0. 905).② rs12979860 polymorphisms was not significantly asso-ciated with the susceptibility to HCV infection in Xinjiang Uyghur populition. Conclusion The IL-28B rs8099917 gene polymorphism correlates with susceptibility to HCV infection in Uyghur patient. G allele is associated with a higher risk of HCV infection.
