乙型肝炎表面抗原阳性孕妇及其新生儿人类TH01基因遗传多态性

摘    要:目的了解乙型肝炎表面抗原(HBsAg)阳性孕妇及其新生儿群体人类(HUM)TH01的遗传多态性,获得这类群体相应多态位点的遗传学数据。方法非抗凝血样采自123对无血缘关系的HBsAg阳性孕妇及其新生儿群体,蛋白酶K法提取模板DNA,聚合酶链反应(PCR)扩增模板DNA后,聚丙烯酰胺凝胶垂直电泳,银染显色分析。结果HBsAg阳性孕妇群体HUMTH01基因座观察到6个等位基因,等位基因频率为TH01*6=0.1138、TH01*7=0.2439、TH01*8=0.0732、TH01*9=0.4472、TH01*9.3=0.0691及TH01*10=0.0610;HBsAg阳性孕妇新生儿观察到6个等位基因,等位基因频率为TH01*6=0.0732、TH01*7=0.2398、TH01*8=0.0894、TH01*9=0.4797、TH01*9.3=0.0813及TH01*10=0.0366。结论HBsAg阳性孕妇及其新生儿群体HUMTH01的等位基因多态性分布均符合哈温(Hardy-Weinberg)平衡定律,且两群体的遗传多态性分布差异无显著性,与中国汉族TH01等位基因分布差异无显著性。

Abstract:Objective The aim of this study on the genetic polymorphiam at TH01 locus was to understand the genetic relationship between HBsAg positive pregnant women and their newborns. Methods Unanticoagulant blood specimens were collected from 123 pairs of unrelated HBsAg-positive pregnant women and their newborns in Taiyuan, Shanxi province. The DNA samples were extracted using protease-K method and amplified by PCR technique. The PAGE vertical electrophoresis and silver-stain were used for typing the PCR product. Results There were six alleles at TH01 locus in HBsAg-positive mothers. The alleles frequence was TH01*6=0.1138, TH01*7=0.2439, TH01*8=0.0732, TH01*9=0.4472, TH01*9.3=0.0691 and TH01*10=0.0610; For newborn population there were also six alleles at TH01 locus. The alleles frequence was TH01*6=0.0732, TH01*7=0.2398, TH01*8=0.0894, TH01*9=0.4797, TH01*9.3=0.0813, and TH01*10=0.0366. Conclusion The genotype distributions observed in the population are in good agreement with Hardy-Weinberg equilibrium, and there is no difference in the distribution of allele frequencies at TH01 locus between mother and baby populations, not different from that in Han nationality in China as well.
