加味桃红四物汤含药血清对动脉粥样硬化血瘀证模型大鼠血管内皮细胞功能的影响

摘    要:目的:观察加味桃红四物汤含药血清对体外培养的动脉粥样硬化血瘀证大鼠血管内皮细胞(VECs)的保护作用及其机制。方法:制备加味桃红四物汤含药血清及动脉粥样硬化血瘀证模型大鼠,从模型大鼠胸主动脉分离培养VECs,将VECs分为对照组、血瘀血清组、中药血清组分别进行处理,采用改良的Boyden chamber法观察VECs迁移的活性,硝酸还原酶法测定培养液中NO含量,ELASA法测定培养液中ET、t-PA/PAI-1、ICAM-1含量,RT-PCR检测eNOSmRNA、ICAMmRNA的表达。结果:血瘀血清组培养液中NO、t-PA含量低于对照组与中药血清组(P〈0.05),ET、PAI-1、ICAM-1含量高于对照组与中药血清组(P〈0.05);VECs经血瘀血清处理后,迁移活性降低,而经中药血清干预后,VECs迁移活性增加,与血瘀血清组比较,差异有统计学意义(P〈0.05)。血瘀血清组eNOSmRNA表达低于中药血清组,而ICAMmRNA表达高于中药血清组。结论:加味桃红四物汤药物成分可促进动脉粥样硬化血瘀证大鼠VECs迁移,并促进VECs保护性因子表达。

Effect of serum containing modified Taohong Siwu decoction on function of vascular endothelial cells in rat model of atherosclerosis and blood stasis syndrome

Abstract:Objective:To observe the protective effect of serum containing modified Taohong Siwu decoction on vascular endothelial cells (VECs) cultured in vitro in rats with atherosclerosis and blood stasis syndrome and its action mechanism. Methods:Serum contai- ning modified Taohong Siwu decoction was prepared, and a rat model of atherosclerosis and blood stasis syndrome was established. VECs were isolated from the thoracic aorta of model rats. VECs were divided into control group, blood stasis serum group, and traditional Chi- nese medicine (TCM) serum group to receive respective treatments. The migration of VECs was observed by the improved Boyden chamber method;the nitric oxide (NO) content of culture solution was determined by the nitrate reductase method;the content of endo- thelin (ET) ,tissue plasminogen activator (t - PA)/plasminogen activator inhibitor - 1 (PAl - 1) ,and intercellular adhesion molecule - 1 ( ICAM - 1 ) in cuhure solution was determined by enzyme - linked immunosorbent assay ; the expression of endothelial nitric oxide synthase (eNOS) mRNA and intercellular adhesion molecule (ICAM) mRNA was measured by RT -PCR. Results:Compared with the control group and TCM serum group, the blood stasis serum group had a significantly lower content of NO and t - PA (P 〈 0. 05 ) but a significantly higher content of ET, PAI- 1, and ICAM - 1 ( P 〈 0. 05 ). VECs had reduced migration after being treated with blood stasis serum, but had increased migration after being treated with TCM serum ; migration showed a significant difference between the two groups (P 〈0.05 ). Compared with the TCM serum group,the blood stasis serum group had a significantly reduced eNOS mRNA expressionbut a significantly increased ICAM mRNA expression. Conclusion : Modified Taohong Siwu decoction can promote the migration of VECs and the expression of protective factors for VECs in rats with atherosclerosis and blood stasis syndrome.
