雷公藤红素对HMC-1细胞凋亡相关基因表达的影响

摘    要:目的:探讨雷公藤红素诱导人肥大细胞系HMC-1细胞凋亡的基因调节机制。方法:雷公藤红素(0.5umol/L)与HMC-1细胞共育不同时间(4、8或12h)后,用RT-PCR、免疫组化方法检测bcl-2家庭、c-myc、白细胞介素1β转换酶(ICE)基因在mRNA和蛋白质水平表达的变化,并分析它们在细胞凋亡中的作用。结果:HMC-1细胞本已存在Bcl-2、Bax、C-myc蛋白表达;雷公藤红素作用后,Bcl-2蛋白表达受到抑制,Bax、C-myc蛋白表达增加。HMC-1细胞能自发表达bcl-2、bax、bcl-XL和ICE的mRNA,在雷公藤红素作用下,bcl-2、bax、bcl-XL的mRNA表达水平均下降(以bcl-2下降最为明显),而ICE mRNA3次检测2次上升,1次下降。结论:雷公藤红素诱导HMC-1细胞凋亡可能与其上调促凋亡基因和下调抑凋亡基因的表达有关。

Abstract:Objective: To explore the molecular mechanism of HMC-1 cell apoptosis on exposure to tripterine. Methods: After the HMC-1 cells were incubated with tripterine, the expression of bcl-2, bax, bcl-X,c-myc and ICE were assayed by using immunohistochemical staining and RT-PCR. Results: The expression of bax,c-myc were up-regulated and bcl-2 down-regulated at protein level.The expression of bax,bcl-X L, especially bcl-2 were down-regulated, and ICE was up-regulated at mRNA level. Conclusion: These results suggest that apoptosis of HMC-1 cells induced by tripterine is regulated by different expression of apoptosis-related genes.
