培养人脐动脉内皮细胞的组织化学和免疫细胞化学的研究

摘    要:用细胞化学方法研究了体外培养人脐动脉内皮细胞的Mg-ATP酶及碱性磷酸酶活性,并与人脐带切片中血管内皮以及大鼠脑、心、肾及肝等切片中血管内皮有关酶相比较。同时,用免疫细胞化学方法检查了培养脐动脉内皮细胞的Ⅷ因子相关抗原。结果显示∶(1)培养48及72h的脐动脉内皮细胞、人脐带血管及大鼠脑、心、肾、肝血管内皮都呈Mg-ATP酶阳性。(2)碱性磷酸酶在上述各标本的血管内皮均呈阴性反应。(3)培养的人脐动脉内皮细胞呈Ⅷ因子相关抗原阳性。以上结果提示∶(1)体外培养人脐动脉内皮细胞和体内重要器官血管内皮的功能有相似性,是一种较为理想的研究人类动脉血管内皮生理功能和疾病的细胞模型。(2)Mg-ATP酶可作为脐血管内皮以及脑、心、肾、肝等器官血管内皮的标志酶之一。

The Cytochemical and Immunocytochemical Studies on Cultured Human Umbilical Artery Endothelial Cells and on Endothelia of Rats in Vivo

Abstract:The activities of Mg ATPase and alkaline phosphatase (AKP) were assayed with the cytochemical method in cultured endothelial cells derived from human umbilical artery (HUAEC) and compared with the activities of the same enzymes in the endothelial cells (EC) of human umbilical cord sections as well as in the brain, heart, kidney and liver sections from rats. The factor Ⅷ related to antigen (fⅧRAg) of HUAEC was examined with the immunocytochemical method. The results were as follows: (1) In HUAEC cultured for 48 and 72 h, and in the ECs of human umbilical cord section as well as in the ECs of brain, heart, kidney and liver from rats the Mg ATPase was positive. (2) The activity of AKP was negative ethier in HUAEC or in ECs mentioned above. (3) The fⅧRAg in HUAEC was positive. The results suggest that: (1) There are some functional similarities between the HUAEC and the ECs of major organs. The HUAEC is an ideal cell model for studying the physiological functions and some diseases in the human artery. (2) The Mg ATPase may be regareded as one of the marker enzymes of HUAEC and the ECs of brain, heart, kidney and liver.
