摘    要:目的 探讨多药耐药性逆转的新方法。方法 应用基因重组方法构建能转录出mdr1 mRNA反义RNA的逆转录病毒质粒,然后将该质粒导入膀胱癌耐药细胞系中,测定转染细胞对不同药物的耐药性,观察反义核酸对MDR的逆转效应。结果 耐药细胞经导入反义核酸逆转录病毒质粒后,对阿霉素(ADM)、秋水仙素(COL)的IC50显著降低(P〈0.01);细胞膜上P-糖蛋白(P-GP)染色信号显著减弱。结论 反义核酸是逆

Abstract:Objective To explore the new reversal method of multidrug resistance of bladder cancer cell line. Methods A retroviral plasmid pLXSN AS( ) containing the mdr1 mRNA antisense gene was cloned by inserting 1383bp fragment of mdr1 cDNA into multiple cloning sites of retroviral vector pLXSN. For evaluating the reversal efficiency of antisense RNA the recombinant plasmid pLXSN AS( ) was transfected into multidrug resistant cell lines BHA 60 with liposome. Results After the recombinant plasmid pLXSN AS( ) being transduced, the BHA 60 cell line showed significant decrease of IC 50 to drug ( P < 0.01) and the P GP positive signal. Conclusions Antisense is a valuable method to reverse multidrug resistance of bladder cancer cell line.
