摘    要:
目的:探讨真核细胞翻译起始因子3a(eukaryotic translation initiation factor 3,subunit A,eIF3a)和人附睾蛋白 4(human epididymis protein 4,HE4)与卵巢癌发生、发展的关系及eIF3a和HE4表达之间的相关性。方法:分别采用RTPCR 和免疫组织化学法检测181例卵巢癌患者、卵巢良性肿瘤患者及正常卵巢组织中eIF3a与HE4的mRNA和蛋白表达 情况。结果:eIF3a基因和HE4基因在正常卵巢、卵巢良性肿瘤及卵巢癌组织中的mRNA及蛋白表达水平均差异有统 计学意义(P<0.05)。eIF3a基因和HE4基因mRNA表达在卵巢癌组织与正常卵巢组织、卵巢良性肿瘤组织与正常卵巢组 织中差异均有统计学意义(P<0.05);其中正常卵巢组织eIF3a基因mRNA表达水平高于卵巢良性肿瘤和卵巢癌组织,而 HE4基因mRNA表达水平从正常卵巢、卵巢良性肿瘤到卵巢癌组织中逐渐升高;HE4基因mRNA表达水平卵巢癌高于 卵巢良性肿瘤组织(P<0.001)。eIF3a蛋白在正常卵巢、卵巢良性肿瘤及卵巢癌组织中的阳性表达率分别为0,66.7% 和 81.0%,HE4蛋白在正常卵巢组织、卵巢良性肿瘤组织及卵巢癌组织中的阳性表达率分别为0,27.8%和56.2%。eIF3a 蛋白和HE4蛋白表达阳性率在卵巢癌组织与卵巢良性肿瘤组织、卵巢癌组织与正常卵巢组织、良性肿瘤组织与正常 卵巢组织中差异均有统计学意义(P<0.001)。eIF3a蛋白和HE4蛋白表达水平呈正相关(r=0.35,P<0.05)。结论:eIF3a和 HE4的表达与卵巢癌发生相关,二者可能通过细胞外调节蛋白激酶通路相互联系。

Abstract:
Objective: To investigate the correlation between eukaryotic translation initiation factor 3, subunitA (eIF3a) and human epididymis protein 4 (HE4) expression and ovarian cancer.Methods: RT-PCR or immunohistochemistry was used to examine eIF3a and HE4 mRNA orprotein expression in ovarian tissues from patients with ovarian cancer (n=181) or benign ovariantumors, or from the healthy women.Results: There were significant differences in mRNA and protein expression of eIF3a and HE4 amongnormal ovarian tissues, benign ovarian tumor tissues, and ovarian cancer tissues (P<0.05). There weresignificant differences in mRNA expression of eIF3a and HE4 between the normal tissues and theovarian cancer tissues, or between the benign ovarian tumor tissues and the normal tissues (P<0.001).The mRNA expression of eIF3a in the normal ovarian tissues was significantly higher than that in thebenign ovarian tumor tissues or that in the ovarian cancer tissues. The mRNA expression of HE4 wasgradually increased from the normal ovarian tissues, the benign ovarian tumor tissues to the ovariancancer tissues. The mRNA expression of HE4 in the ovarian cancer tissues was significantly higherthan that in the benign ovarian tumor tissues (P<0.001). Positive expression rates for eIF3a or HE4protein in normal, benign tumor, and cancer tissues were 0, 66.7%, and 81.0% or 0, 27.8%, and 56.2%,respectively. There were significant differences in positive expression rates of eIF3a protein and HE4protein between the ovarian tumor tissues and benign ovarian tumor tissues, between the ovariancancer tissues and the normal ovarian tissues, or between the benign ovarian tumor tissues and thenormal ovarian tissues (P<0.001). The eIF3a protein expression was positively correlated with HE4protein expression (r=0.575, P<0.05).Conclusion: The expressions of eIF3a and HE4 are associated with ovarian cancer, andextracellular regulated protein kinases may play a role in the interaction between eIF3a and HE4.
