赤芍、白芍凉血作用比较研究(Ⅱ)——抑制脂多糖诱导大鼠肺泡巨噬细胞凋亡的谱效分析

摘    要:目的:以LC-MS图谱上的峰面积和川赤芍、赤芍、白芍对脂多糖(LPS)刺激的大鼠肺泡巨噬细胞NR8383细胞活性的影响为基础进行谱效分析,以此确定川赤芍、赤芍、白芍抑制NR8383细胞凋亡的“药效成分组”,并比较差异.方法:采用偏最小二乘回归法( PLSR),对川赤芍、赤芍、白芍LC-MS图谱数据和NR8383细胞凋亡药效数据之间的内在联系进行谱效分析.结果:LC-MS图谱上的X8,X10,X3,X7,X13这5个成分组成的“药效成分组”对川赤芍、赤芍、白芍抑制NR8383细胞凋亡药效的贡献较大.结论:“药效成分组”的研究能够较为全面的反映中药的功效,是评价中药质量的关键.

Abstract:Objective:To study the fingerprints correlated with pharmacological effect on the basis of LC-MS fingerprint and data of Radix Paeoniae Alba and Radix Paeoniae Rubra,in order to definite the effective compounds of decreasing apoptosis induced by lipopolysaccharide in the alveolar macrophages.Method:The inherent connections were studied between LC-MS fingerprint and decreasing apoptosis of Radix Paeoniae Alba and Radix Paeoniae Rubra,which was induced by lipopolysaccharide in the alveolar macrophages,with partial least squares regression(PLSR).Result:Effective compounds consist of five peaks X8,X10,X3,X7,X13of the fingerprint play the most important role in the decreasing apoptosis induced by lipopolysaccharide in the alveolar macrophages.Conclusion:The ’effective compounds’ are the key of quality evaluation of traditional Chinese medicine and can provide comparatively comprehensive information of Chinese medicine.
