血小板衍生生长因子—BB对血管内皮细胞、平滑肌细胞及成纤维细胞增殖的影响

摘    要:目的 观察血小板衍生生长因子—BB对培养的人血管内皮细胞、兔平滑肌细胞和人成纤维细胞增殖的影响。方法 采用培养的人脐静脉血管内皮细胞、兔动脉血管平滑肌细胞和人血管成纤维细胞,应用^3H—TdR掺入方法,观察血小板衍生生长因子—BB对三种细胞DNA合成的影响。结果 血小板衍生生长因子—BB可促进处于静止状态的三种细胞DNA的合成,并呈现出明显的浓度依赖关系,在30ng/ml的浓度时成纤维细胞DNA的合成达到高峰,在40ng/ml的浓度时内皮细胞、平滑肌细胞DNA的合成达到高峰。成纤维细胞、平滑肌细胞分别在PDCF-BB作用24h和36h年到DNA合成的高峰,内皮细胞在48h时DNA合成量最高。结论 血小板衍生生长因子—BB可明显促进培养的人脐静脉血管内皮细胞、兔动脉血管平滑肌细胞和人血管成纤维细胞的增殖。

Abstract:Objective To investigate the effects of platelet-derived growth factor BB (PDGF-BB)on proliferation of human umbilical vein endothelial cells, rabbit vascular smooth muscle cells and human vascular fibroblasts. Methods The human umbilical vein endothelial cells, rabbit vascular smooth muscle cells and human vascular fibroblasts were cultured. Effects of PDGF-BB on DNA synthesis of the three kinds of cells were studied using 3H-TdR incorporation in vitro. Results PDGF-BB significantly accentuated DNA synthesis of quiescent human vascular fibroblasts, rabbit vascular smooth muscle cells and human umbilical vein endothelial cells in a dose-dependent manner, with a maximal response of fibroblasts at a concentration of 30 ng/ml and vascular smooth muscle cells and endothelial cells at a concentration of 40 ng/ml. The maximal response was induced by a concentration of 30 ng/ml at 24 h, 36 h and 48 h respectively. Conclusion PDGF-BB promote proliferation of cultured human umbilical vein endothelial cells, rabbit vascular smooth muscle cells and human vascular fibroblasts.
