不同胃病组织中多种相关基因表达和幽门螺杆菌感染的研究

目的:探讨不同胃病组织中C-myc、p16、p53、K-ras和C-erbB-2基因表达与幽门螺杆菌(HP)感染的临床意义.方法:同顾性分析胃镜检奁活检标本162例,观察胃黏膜的炎症、上皮化生、上皮内瘤变变化及HP(HP IgG).应用免疫组化SP法检测C-myc、p16、p53、K-ras和C-erb B-2基因的表达.结果:在不同胃病组织中,C-myc、P16、P53基因的表达差异均有显著性.而K-ras和C-erbB-2基因表达差异无显著性.肠上皮化生和上皮内瘤变检出率依非萎缩性胃炎、萎缩性胃炎和胃溃疡顺序逐渐增加.HP感染率以胃癌最高,而在胃溃疡、非萎缩性胃炎和萎缩性胃炎组织中检出率较低.结论:胃癌组织中癌基因表达显著高于非癌变胃病,HP感染与胃癌发生有关.

Expression of C-myc,p16,p53,K-ras and C-erb B-2 and helicobacter pylori infection in different gastropathy tissues

Objective:To explore the clinical significance of helicobacter pylori infection and expression of C-myc,p16,p53,K-ras and C-erb B-2 in different gastropathy tissues.Methods:162 biopsy specimens from gastric mucosa were analysed.The inflammation and dysplasia of gastric mucosa were detected by HE staining,intestinal metaplasia was observed by particular staining,and helicobacter pylori was examined by Warthin-stary silver staining.Serum HP IgG was detected by ELISA.Helicobacter pylori was analysed by 14C-UBT.The expression of C-myc,p16,p53,K-ras and C-erb B-2 was observed by SP method.Results:In different gastric tissues,there were significant differences of C-myc,p16 and p53 expression,but there were no differences of k-ras and C-erb B-2 expression.The incidences of intestinal metaplasia and atypical hyperplasia were increasing with non-atrophic gastritis,atrophic gastritis and gastric ulcer.The rate of HP infection was the most in gastric cancer,and that was lower in other gastriopathies.Conclusion:Our study suggested that expression of oncogens in gastric cancer tissues was higher than that in non-cancerous gastropathy and there was the relationship between HP infection and gastric carcinoma.
