参灵方防治佐剂性关节炎大鼠药源性胃黏膜损伤及对前列腺素E2含量的影响

摘    要:目的探讨参灵方防治佐剂性关节炎(AA)大鼠药源性胃黏膜损伤及对前列腺素(PG)E2含量影响。方法从70只Wistar大鼠中随机取出10只作为空白对照组,其余60只均制作AA大鼠模型,将出现二次反应的大鼠随机取出40只,并随机分为4组,即模型对照组、雷尼替丁对照组及参灵方小、大剂量组,各10只。雷尼替丁对照组予盐酸雷尼替丁胶囊30 mg/(kg.d)灌胃,参灵方小、大剂量组分别予参灵方3.5、7.0 g/(kg.d)灌胃,空白对照组、模型对照组分别予等容积0.9%氯化钠注射液灌胃。灌胃时间均为28 d,于末次灌胃结束后模型对照组与各给药组均采用腹部皮下注射吲哚美辛进一步诱导药源性胃黏膜损伤的复合模型,计算胃黏膜损伤指数、损伤抑制率,测定胃黏膜组织及血清中PGE2含量。结果雷尼替丁对照组及参灵方大、小剂量组胃黏膜损伤指数均较模型对照组减少(P<0.01);参灵方大、小剂量组胃黏膜损伤指数比较差异有统计学意义(P<0.05)。空白对照组大鼠胃黏膜色泽红润,未见损伤性改变;模型对照组大鼠胃黏膜表面可见点线状出血,少数呈斑片状;各用药组均仅见针尖样出血及点状糜烂,其中以雷尼替丁组及参灵方小剂量组较好,参灵方大剂量组次之。与空白对照组比较,模型对照组胃黏膜组织及血清中PGE2含量均降低(P<0.05);与模型对照组比较,雷尼替丁对照组及参灵方大、小剂量组胃黏膜组织及血清中PGE2含量均升高(P<0.05)。结论参灵方对AA大鼠药源性胃黏膜损伤有保护作用。

Effects of Shengling decoction on the levels of prostaglandin E2 and drug - induced gastric mucosal injury in AA rats

Abstract:Objective To explore the effect of Shengling decoction on the levels of prostaglandin E2 and drug -induced gastric mucosal injury in adjuvant arthritis (AA) rats. Methods 70 Wistar rats were randomly divided into 10 rats in control group and 60 rats to be Adjuvant arthritis (AA) models. 40 rats that presenting secondary re- sponse in 60 rats model were divided randomly into model group, ranitidine group, Shengling decoction low - dose group and Shengling decoction high -dose group. Rats were given corresponding drug once a day and keep 28 days [ ranitidine group 30 mg/( kg ~ d) ; Shengling decoction each group ,3.5,7 g/( kg ~ d) ]. The gastric mucosal mod- els were established on the basis of AA models by subcutaneous injecting Indomethacin. The gastric mueosa lesion index was observed and damage inhibitory rate and the prostaglandin E2 content in gastric mucosa and serum were examined. Results Compared with model group, the gastric mucosa lesion index in ranitidine group, Shengling decoction low - dose group and Shengling decoction high - dose group were decreased ( P 〈 0. 01 ). There was significant difference between Shengling decoction low - dose group and Shengling decoction high - dose group on the gastric mu- cosa lesion index ( P 〈0. 05 ). Gastric mucosa color is red and no traumatic change in control group. There was visi- ble point splinter hemorrhages in the gastric mueosa surface and a few patchy in the model group. There was only pin- point bleeding and punctate erosion in each treatment group. Compared with control group, the levels of PGE2 in ser- um and gastric mucosa tissue in model group was decreased ( P 〈 0.05 ). Compared with model group, the levels of PGE2 in serum and gastric mucosa tissue in ranitidine group, Shengling decoction low - dose group and Shengling de- coction high - dose group were decreased ( P 〈 0.05 ). Conclusion s Shengling decoction has very good protection effect on druginduced gastric mucosal injury in AA rats.
