摘    要:目的:研究长链非编码RNA表观遗传诱发长链非编码RNA1(epigenetically induced long non-coding RNA 1, EPIC1)在肝细胞肝癌(hepatocellular carcinoma, HCC)中的表达情况及临床价值。方法:收集2013年9月—2018年3月被确诊为HCC患者48例的肿瘤及癌旁组织,通过实时荧光定量聚合酶链式反应(quantitative real-time polymerase chain reaction, qRT-PCR)检测HCC组织、细胞中EPIC1的表达情况;分析EPIC1表达与患者临床参数的相关性。结果:EPIC1在肝癌组织中平均表达量为(13.28±36.66),显著低于癌旁组织(62.84±108.93)(P=0.006)。相较于正常的肝细胞系L-02,EPIC1在HCC细胞系中低表达(P<0.05)。HCC患者EPIC1表达量与患者的TNM分期相关(P=0.033)。结论:EPIC1在HCC中低表达,可能为潜在的治疗靶点。

Abstract:Objective: To determine the expression level and clinical value of long non-coding RNA epigenetically induced long non-coding RNA 1(EPIC1) in hepatocellular carcinoma(HCC). Methods: A total of 48 paired tissues were collected from HCC patients between September 2013 and March 2018. The relative expression level of EPIC1 in tissues and cell lines were detected by quantitative real-time polymerase chain reaction(qRT-PCR) assays. Then the correlation of the expression of EPIC1 with clinical features in HCC was analyzed. Results: The EPIC1 expression level was significantly down-regulated in the HCC tissues(13.28±36.66) compared with that in the adjacent normal tissues(62.84±108.93), and the difference was statistically significant(P=0.006). The expression levels of EPIC1 in HCC cell lines were significantly lower than that in normal human hepatocyte cell line(L-02)(P<0.05). Low levels of EPIC1 were correlated with advanced TNM stage(P=0.033). Conclusion: EPIC1 is down-regulated in HCC tissues and cell lines and it may be a potential therapeutic target in the future.
