脐带问充质干细胞对兔眼碱烧伤的治疗研究

摘    要:
目的探讨脐带间充质干细胞(UC—MSCs)的甲基纤维素涂剂对兔角膜损伤的修复作用。方法体外进行UC—MSCs的培养和鉴定,采用NaOH溶液制作兔角膜烧伤模型.烧伤后以甲基纤维素为载体将UC—MSCs移植到烧伤的兔角膜表面.以单纯甲基纤维素组及碱烧伤组作为对照组,在裂隙灯下观察角膜的临床改变。结果成功地培养出UC-MSCs;碱烧伤阳性对照组和单纯甲基纤维素组均可见角膜浑浊呈瓷白色、表面粗糙,不能透见眼内结构。UC—MSCs甲基纤维素组,新生血管较少。角膜表面较光滑.角膜透明度明显改善。结论UC—MSCs可以在兔角膜表面成活并向角膜上皮细胞分化,UC-MSCs的甲基纤维素徐剂对碱烧伤的角膜损伤有一定的修复作用。

Abstract:
Objective To explore the repair effect of the umbilical cord mesenchyma stem cell (UC-MSCs) methylcellulose medicinal preparation to the injury of rabbit cornea. Methods UC-MSCs were cultivated and identified in vitro. Rabbit cornea burn model was made using NaOH solution. After the burn, UC-MSCs were transplanted to the burn rabbit cornea surface, taking methylcellulose as carrier. Pure methylcellulose group and alkali burn group were control group. The clinical change of the cornea was observed under the slit lamp. Results UC-MSCs were cultivated successfully. In the alkali burn masculine control group and the pure methylcellulose group, corneas were polluted and porcelain white, the surfaces were rough, and the intraocular structure could not be seen. In UC-MSCs methylcellulose group, the newborn blood vessels were few, the cornea surfaces were smooth, cornea transparency was obviously improved. Conclusions UC-MSCs may survive in the rabbit cornea and differentiate to the epithelia corneal cell, the UC-MSCs methylcellulose has certain repair effect to cornea injury of alkali burn.
