摘    要:
本文用硫代巴比妥酸法(TBA)和DPH 荧光偏振技术探讨了V_2O_5对大鼠肾脏毒性的机理。结果表明V_2O_5在体内、外均能诱发大鼠肾皮质细胞生物糢的脂质过氧化和降低膜脂流动性,并呈现明显的时间-效应和剂量-效应关系。体外染毒后的作用部位主要在线粒体和微粒体,而体内染毒后,主要限于微粒体。醋酸锌和EDTA 在体外实验中能抑制V_2O_5所诱发的脂质过氧化和膜脂流动性的改变。结果还提示,膜脂过氧化与膜脂流动性之间存在依赖关系。

Abstract:
In this study the mechanisms of nephrotoxicity of V_2O_5were investi-gated with TBA and DPH fluorscence polarization.The results showedthat V_2O_5was able to induce lipid peroxidaton and decrease membrane fl-uidity in biological membrane of renal cortex in rats,chiefly affecting those or mitochondria and microsomes in vitro and merely those ofmicrosomes in vivo,both showed significant dose-dependent and timedependent relationships.Zinc acctatc and EDTA displayed separately invitro,the capability of inhibiting V_2O_5-induced lipid peroxidation andchanges in membrane fluidity.These results suggested that there existdependent relationship between lipid peroxidation and membrane fluidity.
