缺氧促进牛视网膜血管内皮细胞VEGF的表达

摘    要:目的探讨缺氧对视网膜血管内皮细胞VEGF表达的影响。方法采用细胞克隆、机械除杂法培养视网膜血管内皮细胞;四唑盐比色试验(MTT)比较正常和缺氧状态下牛视网膜血管内皮细胞的细胞数目。应用ELISA法检测正常和缺氧状态下牛视网膜血管内皮细胞VEGF的表达。缺氧状态下加入VEGF抗体后,检测VEGF的表达。结果正常状态下即可检测到VEGF的表达。缺氧状态下,牛视网膜血管内皮细胞VEGF的表达显著增加(P<0.01)。结论缺氧环境能刺激培养的牛视网膜血管内皮细胞的增殖。缺氧能刺激培养的牛视网膜血管内皮细胞VEGF的表达。

Abstract:Objective] To investigate expression of vascular endothelium growth factor (VEGF) of bovine retina microvascular endothelial cell under hypoxic condition. Method] Bovine retina microvascular endothelial cell were obtained by using cell cloning separating and pericyte weeding. By using MTT, we described cell number of retina microvascular endothelial cell under normal and hypoxic condition. By using ELISA method, we explored the expression of VEGF under normal and hypoxic condition. Detected the expression of VEGF after adding anti-VEGF under hypoxia. Result] VEGF could be detected under normal condition. Under hypoxic ndition, the expression of VEGF was increased obviously. Conclusion] Hypoia can stimulate proliferation of endothelial cell. Hypoia can stimulate expression of VEGF in bovine retina microvascular endothelial cell.
