摘    要:目的 探讨半乳糖凝集素-9(Galectin-9)对卵巢癌细胞侵袭转移的影响.方法 将不同浓度外源性Galectin-9[0(阴性对照)、4μg/mL、8μg/mL、16μg/mL]加入卵巢癌SKOV3细胞中,培养48 h,Western blot法检测细胞内Galectin-9的表达,MTT法检测细胞活力,Transwell法检测细胞迁移及侵袭能力,Western blot检测基质金属蛋白酶2(MMP2)、MMP9、上皮间充质转化(EMT)相关蛋白E-钙粘蛋白(E-cadherin)及波形蛋白(Vimentin)的表达.结果 与阴性对照比较,4μg/mL、8μg/mL、16μg/mL外源性Galectin-9能显著提高卵巢癌SKOV3细胞中Galectin-9的表达(P<0.01);4μg/mL、8μg/ml、16μg/mL外源性Galectin-9能显著降低SKOV3细胞活力[(100.00±7.46)vs(77.92±8.03)、(60.62±6.42)、(47.63±5.41)](P<0.01),减少细胞迁移[(99.49±9.63)vs(78.36±8.90)、(65.42±7.32)、(50.31±6.78)]及侵袭数目[(99.13±10.30)vs(76.69±7.23)、(63.68±6.32)、(52.37±4.99)](P<0.01),并能显著下调MMP2、MMP9及Vimentin的表达(P<0.01),上调E-cadherin的表达(P<0.01).结论 上调卵巢癌细胞SKOV3中Galectin-9的表达能显著抑制细胞迁移侵袭等恶性生物学行为,可能与下调MMPs及调节EMT相关蛋白表达有关.

Effect of Galectin-9 on invasion and metastasis of ovarian cancer cells SKOV3
Abstract:Objective To explore effect of Galactose lectin-9 (Galectin-9) on invasion and metastasis in ovarian cancer cells SKOV3. Methods SKOV3 cells were treated with different concentrations of exogenous Galectin-9 [0 (neg-ative control), 4μg/mL, 8μg/mL, 16μg/mL] for 48 h. The expression of Galectin-9 in SKOV3 cells was detected by West-ern blot. Cell viability was measured by MTT assay, and the ability of cell migration and invasion was detected by tran-swell assay. The expression of matrix metalloproteinase 2 (MMP2), MMP9, EMT related proteins E-cadherin and Vimentin was detected by Western blot. Results Compared with negative control, 4μg/mL, 8μg/mL, 16μg/mL Galectin-9 upreg-ulated the expression of Galectin-9 in SKOV3 cells (P<0.01), and inhibited SKOV3 cells viability [(100.00 ± 7.46) vs (77.92 ± 8.03), (60.62 ± 6.42), (47.63 ± 5.41), P<0.01], migration [(99.49 ± 9.63) vs (78.36 ± 8.90), (65.42 ± 7.32), (50.31 ± 6.78), P<0.01] and invasion [(99.13±10.30) vs (76.69±7.23), (63.68±6.32), (52.37±4.99), P<0.01], as well as down-regulat-ed the expression of MMP2, MMP9 and Vimentin (P<0.01) and upregulated the expression of E-cadherin (P<0.01). Conclusion Overexpresion of Galectin-9 in SKOV3 cell could inhibit cell migration, invasion and metastasis, which is associated with down-regulation of the expression of MMPs and regulation of the expression of EMT related proteins.
