宫颈癌SiHa细胞和卵巢癌SKOV3细胞染色体着丝粒点变异的研究

摘    要:目的:探讨染色体着丝粒点(Cd)的变异与宫颈癌S iHa细胞和卵巢癌SKOV3细胞染色体非整倍性畸变的关系。方法:肿瘤细胞株传代后用常规方法制备染色体,用直接法制备胚胎绒毛组织染色体标本,采用Cd-NOR同步银染分析技术研究S iHa细胞和SKOV3细胞染色体Cd的变异。结果:S iHa细胞染色体Cd缺失率为2.70%、Cd迟滞复制率为0.92%、小Cd率为1.58%、Cd-NOR融合率为0.54%、与正常人胚胎绒毛细胞染色体Cd相比较,S iHa细胞染色体Cd缺失显著升高(P<0.0 1 2 5)、Cd迟滞复制显著升高(P<0.0125)。而小Cd、Cd-NOR融合没有显著性差异。SKOV3细胞染色体Cd缺失率为1.95%、Cd迟滞复制率为0.22%、小Cd率为1.62%、Cd-NOR融合率为1.65%、与正常人胚胎绒毛细胞染色体Cd相比较,SKOV3细胞染色体Cd缺失显著升高(P<0.0125)、Cd-NOR融合显著升高(P<0.0125),而Cd迟滞复制、小Cd没有显著性差异。结论:S iHa细胞染色体非整倍性畸变可能主要源于Cd缺失、Cd迟滞复制。SKOV3细胞染色体非整倍性畸变可能主要源于Cd缺失、Cd-NOR融合。

Abstract:Objective:To study the relation between the centromeric dots(Cd)variation of SiHa cells and SKOV3 cells and the chromosome aneuploidy aberration.Methods: Chromosome of tumor cells were made with general method,Chromosome of normal embryonic villi were made straightly.A Cd-NOR in-phase argentum dye analysis technique modified by this laboratory was used to study centromeric dots(Cd) variation of SiHa cells and SKOv3 cells.Results: Frequency of Cd loss in SiHa cells was 2.70%,Frequency of Cd duplication laggard was 0.92%,frequency of little Cd was 1.58%,frequency of Cd-NOR amalgamation was 0.54%.Frequency of Cd loss and frequency of Cd duplication laggard in SiHa cells were significantly higher than those of normal embryonic villi(P<0.0125).Frequency of Cd-NOR amalgamation and frequency of little Cd showed no difference between SiHa cells and normal embryonic villi.Frequency of Cd loss in SKOV3 cells was 1.95%,frequency of Cd duplication laggard was 0.22%,frequency of little Cd was 1.62%,frequency of Cd-NOR amalgamation was 1.65%.Frequency of Cd loss and Frequency of Cd-NOR amalgamation in SKOV3 cells were significantly higher than those of normal embryonic villi(P<0.0125).Frequency of Cd duplication laggard and frequency of little Cd showed no difference between SKOV3 cells and normal embryonic villi.Conclusion: Cd loss and Cd duplication laggard might be related with aneuploidy aberration of SiHa cells.Cd loss and Cd-NOR amalgamation might be related with aneuploidy aberration of SKOV3 cells.
