当归化学成分的分离与鉴定

摘    要:目的研究当归Angelica sinensis(Oliv.)Diels的化学成分,为其药理作用的研究奠定物质基础。方法采用硅胶柱色谱及制备薄层色谱等方法进行分离纯化,根据理化性质和波谱数据(1H-NMR、13C-NMR等)鉴定化合物的结构。结果分离得到13个化合物,分别鉴定为:正丁基苯酞(3-butylphthalide,1)、4羟基3丁基苯酞(4-hydroxy-3-butylphthalide,2)、2-methoxy-2-(4'-hydroxyphenyl)ethanol(3)、反式阿魏酸(ferulic acid,4)、厚朴酚(magnolol,5)、黄芩苷(baicalin,6)、2″O(2甲基丁酰基)异当药黄素[2″O(2-methylbutyryl)-isoswertisin,7]、harman(8)、1-ace-tyl-β-carboline(9)、酒渣碱(flazine,10)、菠菜甾醇(α-spinasterol,11)、异欧前胡素(isoimperatorin,12)、发卡二醇[3(R),8(S)-falcarindiol,13]。结论化合物3、7~9、11为首次从该属植物中分离得到,化合物5为首次从该植物中分离得到。

Abstract:Objective To isolate and identify the chemical constituents from roots of Angelica sinensis (Oliv.) Diels. Methods Compounds were isolated by various kinds of column chromatography methods and preparative TLC, and their structures were identified by the physico-chemical characters and the spectral analysis. Results Thirteen compounds were isolated and their structures were identified as: 3-butylphthalide (1), 4-hydroxy-3-butylphthalide (2), 2-methoxy-2-(4′-hydroxyphenyl)ethanol (3), ferulic acid (4), magnolol (5), baicalin (6), 2²-O-(2¢²-methylbutyryl)-isoswertisin (7), harman (8), 1-acetyl-β-carboline (9), flazine (10), α-spinasterol (11), isoimperaterin (12) and 3(R), 8(S)-falcarindiol (13). Conclusion Compounds 3, 7-9, 11 are isolated from Angelica genus for the first time, and compound 5 is isolated from this plant for the first time.
