M型钾离子通道开放剂对缺血缺氧性脑病的保护作用

摘    要:目的 探讨M型钾离子通道开放剂对缺血缺氧性脑病的保护作用.方法 健康Wistar大鼠15只,采用随机数表法分为假手术组、脑病组和干预组,每组5只.于建模后6 h、12 h、24 h、48 h、72 h抽取每组大鼠尾部的外周静脉血,检测神经元特异性烯纯化酶(NSE)、髓鞘碱性蛋白(MBP)、S-100β蛋白、磷酸肌酸激酶脑型同工酶(CK-BB)、超氧化物歧化酶(SOD)和基质金属蛋白酶9(MMP-9).结果 脑病组和干预组随建模时间延长,血清NSE、MBP、S-100β蛋白、CK-BB、MMP-9先升高后降低,最高点在24 h.血清SOD先降低后升高,最低点在24 h;脑病组在建模后不同时间点的血清NSE、MBP、S-100β蛋白、CK-BB、MMP-9高于假手术组和干预组,而血清SOD低于假手术组和干预组,差异均有统计学意义(P<0.05);干预组在建模后不同时间点的血清NSE、MBP、S-100β蛋白、CK-BB、MMP-9高于假手术组,而血清SOD低于假手术组,差异均有统计学意义(P<0.05).结论 M型钾离子通道开放剂对缺血缺氧性脑病具有良好的保护作用.

Abstract:Objective To investigate the protective effect of M-type potassium channel opener on hypoxic ischemic encephalopathy. Methods A total of 15 healthy Wistar rats were divided into the sham operation group, en-cephalopathy group, intervention group by random number table method, with 5 rats in each group. Peripheral venous blood was extracted from the tail of each rat after modeling 6 h, 12 h, 24 h, 48 h, 72 h. Related factors (Neuron specific enolase [NSE], Myelin basic protein [MBP], Creatine kinase BB [CK-BB], S-100β protein, superoxide dismutase (SOD), matrix metalloproteinase-9 [MMP-9]) were detected. Results As the modeling time extended in the encepha-lopathy group and intervention group, serum NSE, MBP, S-100βprotein, CK-BB, MMP-9 increased first and then de-creased, with the highest point at 24 h. Serum SOD decreased first and then increased, with the lowest point at 24 h. Se-rum NSE, MBP, S-100βprotein, CK-BB, MMP-9 at different time points after modeling (6 h, 12 h, 24 h, 48 h, 72 h) in the encephalopathy group were significantly higher than those in the sham operation group and intervention group, while serum SOD at different time points after modeling (6 h, 12 h, 24 h, 48 h, 72 h) in the encephalopathy group were significantly lower than that in the sham operation group and intervention group (P<0.05). Serum NSE, MBP, S-100βprotein, CK-BB, MMP-9 at different time points after modeling (6 h, 12 h, 24 h, 48 h, 72 h) in the interven-tion group were significantly higher than those in the sham operation group, while serum SOD at different time points after modeling (6 h, 12 h, 24 h, 48 h, 72 h) in the intervention group were significantly lower than that in the sham op-eration group (P<0.05). Conclusion M-type potassium channel opener has a good protective effect on hypoxic isch-emic encephalopathy.
