恩度联合微波消融治疗鼠肝癌移植瘤研究

摘    要:目的 研究恩度联合微波消融治疗小鼠肝癌的效果及其作用机制.方法 将1.0× 107/ml、0.1 mlHepG2肝癌细胞接种到48只荷瘤小鼠的右后腿股部皮下,肿瘤直径约1.5 cm时小鼠随机分成4组.对照组:尾静脉推注0.9%的生理盐水;联合治疗组:尾静脉推注恩度联合微波消融肿瘤;微波消融组:皮下微波消融肿瘤;恩度治疗组:尾静脉推注恩度.对联合治疗组及微波消融组先对皮下肿瘤进行微波消融.治疗第15天每组处死6只荷瘤小鼠,计算抑瘤率,检测血浆VEGF浓度,观察小鼠生存时间.结果 联合治疗组荷瘤小鼠抑瘤率为73.2%,高于其他治疗组(P<0.05);联合治疗组血浆VEGF水平为(31.64±3.64) pg/ml,低于其他治疗组(P<0.05),且小鼠生存时间较其他三组延长,差异有统计学意义(P<0.05).结论 恩度联合微波消融能更好的发挥抑瘤效应,并能延长小鼠总生存期.

Abstract:Objective To study the inhibitory effects and mechanism of endostar combined with microwave ablation in the treatment of transplanted tumor in mice.Methods Forty-eight mice implanted with 0.1 ml HepG2 cells(1.0×107/ml) were randomly divided into four groups:the control group(tail vein injection of 0.9% saline),the combined treatment group(tail vein injection of endostar and microwave ablation),microwave ablation group(tail vein injection of microwave ablation),and endostar group(tail vein injection of endostatar),each with 12 cases.The tumor weight and the inhibition rate were calculated,the expression of vascular endothelial growth factor(VEGF) of tumors was examined,and the survival time was observed.Results The inhibitory rate of the combined treatment group was 73.2%,obviously higher than that of other groups(P<0.05).The expression of VEGF in the combined treatment group was(31.64±3.64) pg/ml,significantly lower than that in other groups(P<0.05).The survival time in the combined treatment group were significantly longer than that in other groups(P<0.05).Conclusion Endostar combined with microwave ablation is preferred for treating tumors and prolonging survival time in mice.
