Caspase-3和Bcl-2蛋白在产时发热胎盘组织中的表达及其与新生儿脑病相关性

目的 研究产时发热患者胎盘组织中Caspase-3和Bcl-2的表达及其与新生儿缺氧缺血性脑病(HIE)的相关性,进一步探讨产时发热导致HIE的可能机制,为其防治提供新路径.方法 收集产时发热患者胎盘组织60例(HIE组30例,非HIE组30例)和正常孕妇组30例,采用免疫组织化学染色法检测胎盘绒毛滋养层Caspase-3和Bcl-2蛋白的表达,通过免疫组化积分判断其表达高低.结果 HIE组胎盘滋养层Caspase-3和Bcl-2表达的免疫组化积分分别为5.05±0.78、1.54±0.43,非HIE组分别为3.03±1.36、3.51±1.59,正常孕妇组分别为2.87±0.68、4.03±0.64.HIE组Caspase-3的表达明显高于非HIE组和正常组(JP＜0.05),而Bcl-2则明显低于非HIE组和正常组(P＜0.05).Caspase-3与Bcl-2在HIE组胎盘组织中的表达呈负相关(r=0.79,P＜0.05).结论 Caspase3在HIE组胎盘组织中的表达增高,Bcl-2的表达降低,两者在胎盘组织中的表达平衡失调可能是HIE发生的原因之一.

Expression of Caspase-3 and Bcl-2 in placenta tissues from prenatal fever patients and their relationship with neonatal hypoxic-ischemic encephalopathy

Objective To determine the expression of Caspase-3 and Bcl-2 in placenta tissues of patients with prenatal fever and its relationship with neonatal hypoxic-ischemic encephalopathy（HIE）.Methods Sixty placenta samples of patients with prenatal fever（HIE-30,non-HIE-30） and 30 normal placenta tissues were enrolled from the third trimester pregnancies.Immunohistochemistry（SP method） was used to study the expression of Caspase-3 and Bcl-2 in human trophoblasts.Results The expression of Caspase-3 in placental trophoblasts was significantly higher in HIE group than in the non-HIE group（5.05±0.78 vs 3.03±1.36,P0.05） and control group（5.05±0.78 vs 2.87±0.68,P0.05）.However,the expression of Bcl-2 was obviously lower in the HIE group than in the non-HIE group（1.54±0.43 vs 3.51±1.59,P0.05） and control group（1.54±0.43 vs 4.03±0.64,P0.05）.The expression of Caspase-3 was inversely correlated with that of Bcl-2 in HIE group.Conclusion The Caspase-3 expression increases and the Bcl-2 expression decreases in HIE group.The imbalance between Caspase-3 and Bcl-2 expression may be one of the causes of HIE.
