Clinical significance of telomerase activity detected in fine-needle aspiration specimens to liver cancer diagnosis
Abstract:AIM To detect the telomerase activity in samples of liver cancer and investigate its clinical significance in liver cancer diagnosis with the fine-needle aspiration technology. METHODS Forty-two liver tissue specimens were obtained by fine-needle aspiration and telomerase activity in hepatecytes was detected with TRAP-ELISA. RESULTS Twenty-five out of 42 cases were verified as liver cancer, 9 as hepatic cirrhosis, 2 as hepatic hemangioma and 6 as chronic hepatitis. In 25 cases, 14 showed positive telomerase activity among 16 fine-needle aspiration diagnosed liver cancers. Four of 5 suspicious positive fine-needle aspiration specimens showed telomerase activity and 2 of 4 fine-needle aspiration negative specimens. The total positive rate of telomerase activity was 80%(20/25). Combined fine-needle aspiration and telomerase activity detection, the confirmed diagnosis rate of liver cancer could be raised from 64% (16/25) to 88% (22/25). in addition, telomerase activity could not be detected in 2 hepatic hemangioma and 6 chronic hepatitis and could only be detected in 3 of 9 hepatic cirrhosis. CONCLUSION The telomerase activity in liver cancer tissues is noticeably higher than in hepatic benign lesions (hepatic cirrhosis, chronic hepatitis, hepatic hemangioma). Combined with fine-needle aspiration, the premature diagnosis rate of liver cancer can be raised and telomersee activity detection is a useful biological index for the clinical application of liver cancer diagnosis.
