依达拉奉预给药对缺血-再灌注损伤大鼠心肌梗死范围及血中TNF-α、IL-1β的影响

摘    要:目的观察依达拉奉对大鼠缺血-再灌注损伤心肌梗塞范围及血中自介素-1β(IL-1β)、肿瘤坏死因子α(TNF-α)的影响。方法实验共设5个组:假手术组、缺血再灌注组(I—R组)和依达拉奉高、中、低剂量组。采用结扎大鼠冠状动脉前降支30min开放120min建立心肌缺血-再灌注损伤(I—RI)模型,通过依达拉奉预处理,观察I—RI大鼠心肌梗死范围以及血中的IL-1β、TNF—α含量的变化。结果依达拉奉高、中两个剂量组梗塞程度明显减轻,梗塞区重占全心脏及左心室的百分比与模型组比较均有显著性差异(P〈0.01~0.001),以依达拉奉中剂量组为优;依达拉奉给药后血中TNF—α、IL-1β有不同程度的下降,其中以高、中剂量组为优(P〈0.01)。结论依达拉奉能够减轻TNF-α、IL—1β对缺血-再灌注心肌的损伤。

Abstract:Objective To observe the effect of edaravone on myocardial infi'action size and IL- 1β, TNF-α in blood in myocardial ischemia reperfusion injury rats. Methods The rats were divided into five groups : sham operation group, I/R group, three treatment groups treated with low, middle and high dosage of edaravone. The model of I/R was induced by ligating the anterior descending branch of the left coronary artery for 30min/opening for 120min. The effect ofedaravone pretreatment on the size of myocardial infarction and the changes of IL-1β and TNF-α in blood were observed. Result The extent of myocardial infarction in groups treated with high and low dosage of edaravone was reduced significantly compared with model group, the percentage of infarcted myocardium weight of the whole heart and left ventricle had significant difference ( P 〈 0.01,0.001), effect of middle dosage edaravone treated group was excellent. IL-1β and TNF-α in blood were down regulated to different degree after giving edaravone, and the effect of high and middle dosage edaravone treated group was excellent ( P 〈 0.01). Conclusion Edaravone could reduce ischemic reperfused myocardium injury caused by IL-1β and TNF-α.
