独活中总香豆素的超临界CO2提取工艺研究

摘    要:目的:优化独活中总香豆素的超临界CO2提取工艺并简单纯化提取液。方法:以提取率和浸膏中总香豆素的含量为指标,通过单因素和正交试验,考察超临界CO2萃取过程中,温度、压力、时间和夹带剂对提取率和含量的影响,确定最优提取工艺。采用干燥法和水萃取法对提取浸膏进行简单纯化处理。结果:综合比较,最优提取工艺为:萃取压力25 mPa,萃取温度50℃,萃取时间4 h。在此条件下,浸膏中总香豆素含量达42.43%,提取率达81.49%。浸膏经干燥处理,总香豆素含量提高到52.35%。用水萃取法,当水温为90℃时,含量可达到54.55%。结论:超临界CO2萃取技术是独活中总香豆素提取的一种高效方法。

Abstract:Objective:To optimize the condition of supercritical fluid extraction(SFE)for total coumarins in radix angelicae pubescentis.Methods:The single factor experinment and orthogonal experiment were adopted to study the effects of factors including pressure,temperature,time and entrainer on extraction rate and content of total coumarins,using drying and water extraction methods to purify the extract.Result:The optimal conditions should be as follows:extraction pressure for 25 mPa,extraction temperature for 50 ℃,extraction time for 4 h.The content of total coumarins in extract reached 42.43% and the extraction rate was 81.49%.The total coumarins content increased to 52.35% when the extract was dried at 70 ℃.The content could reach to 54.55% after water extraction and the water temperature was 90 ℃.Conclusion:Supercritical CO2 extraction technology is an efficient way to extract total coumarins from radix angelicae pubescentis.
