摘    要:目的观察参桂汤对垂体后叶素所致大鼠急性心肌缺血的保护作用。方法取正常SD大鼠56只,随机分为7组,每组8只,分别为空白组,假手术组,模型组,胺碘酮组(25 mg/kg),参桂汤高(3.060 g/200 g)、中(1.530 g/200 g)、低(0.765 g/200 g)剂量组,灌胃相应剂量药物14 d。末次给药1 h后假手术组舌下静脉推注生理盐水0.5 mL/kg,空白组不做处理,其余各组舌下静脉快速推注垂体后叶素0.5 U/kg(1 U/mL)],造成心肌缺血大鼠模型。通过心电图观察各组药物对大鼠心电图ST段的影响,ELISA法检测各组血清中超氧化物歧化酶(SOD)、一氧化氮合成酶(iNOS)、丙二醛(MDA)的活性水平变化。结果与空白组比较,注射垂体后叶素后模型组大鼠心电图ST段明显升高,血清MDA和iNOS活性水平均升高,SOD活性水平显著下降,差异均具有统计学意义(P<0.05或P<0.01);与模型组比较,胺碘酮组、参桂汤高、中剂量组血清MDA和iNOS的活性水平降低,SOD活性水平升高,差异具有统计学意义(P<0.05或P<0.01)。结论参桂汤通过降低急性心肌缺血大鼠血清MDA和iNOS活性水平,提高SOD活性水平,起到保护大鼠急性心肌缺血的作用。

Abstract:Objective:To observe the protective effect of Shengui Decoction on acute myocardial ischemia induced by pituitrin in rats.Methods:56 normal SD rats were randomly divided into 7 groups:the blank group,the sham operation group,the model group,the amiodarone group(25 mg/kg),the Shengui Decoction high(3.060 g/200 g),medium(1.530 g/200 g)and low(0.765 g/200 g)dose groups.The corresponding dosage of drugs were given to the stomach for 14 days.After the last administration of drugs,normal saline(0.5 mL/kg)was injected into the sublingual vein of sham operation group.The blank group were not dealt with any treatments.The other groups were injected with rapid sagging of the sublingual vein.The rat model of myocardial ischemia was induced by 0.5 U/kg(1 U/mL)of somatophyllin.The effects of drugs on ST segment of electrocardiogram were observed by electrocardiogram.The changes of SOD,iNOS and MDA in serum of each group were detected by ELISA.Results:Compared with the blank group,ST segment of ECG and the activity of MDA and iNOS in serum increased significantly and the level of SOD activity decreased significantly in the model group;the differences were statistically significant(P<0.01).Compared with the model group,the activities of MDA and iNOS in the serum of amiodarone group and high and medium dose groups of Shengui Decoction decreased,while the activities of SOD increased;the differences were statistically significant(P<0.01).Conclusion:Shengui Decoction can protect acute myocardial ischemia injury by reducing the activity of serum MDA and iNOS and increasing the activity of SOD in rats with acute myocardial ischemia.
