膝康止痛方对大鼠膝骨关节炎镇痛作用研究

摘    要:目的观察膝康止痛方对膝关节炎的治疗作用及可能机制。方法通过扭体法观察膝康止痛方的镇痛作用;前交叉韧带离断法复制大鼠膝骨关节炎模型,观察膝康止痛方对大鼠膝关节炎的治疗作用,采用HE,Masson,甲苯胺蓝染色方法对膝关节进行病理学观察,ELISA法对大鼠血清中IL-1β和TNF-α含量检测。结果膝康止痛方明显减少醋酸所致的小鼠扭体次数;减轻大鼠膝关节炎的病变程度,降低病理评分,减少大鼠血清中IL-1β和TNF-α的含量。结论膝康止痛方具有明显的镇痛疗效,治疗大鼠膝关节炎,该作用与降低IL-1β和TNF-α有关。

Abstract:Objective To observe the therapeutic effect of Xikang Zhitong decoction on knee osteoarthritis and its possible mechanism.Methods To observe the analgesic effect of Xikang Zhitong decoction by Writhing test.The therapeutic effect of Xikang Zhitong decoction was observed on rat knee osteoarthritis by anterior cruciate ligament dissection.HE,Masson and toluidine blue staining were used for pathological observation of the knee joint.IL-1βand TNF-αin serum of the rats were detected by ELISA.Results Xikang Zhitong decoction reduced the writhing times of mice induced by acetic acid.The pathological degree was reduced of knee osteoarthritis in rats,and the pathological score was decreased.IL-1βand TNF-αwere reduced in serum of knee osteoarthritis rats.Conclusion Xikang Zhitong decoction has obvious analgesic effect and treatment on knee osteoarthritis,which is related to the reduction of IL-1βand TNF-α.
