超高效液相色谱法测定人体血浆中水杨酸含量的方法学研究

摘    要:目的:建立人体血浆中水杨酸含量的超高效液相色谱(ultra performance liquid chromatography,UPLC)测定方法。方法:采用Waters Spursil C18(100 mm×2.10 mm,3μm)色谱柱;乙腈∶水(含0.2%磷酸)=20.7∶79.3为流动相,用前经0.22μm的滤膜过滤,超声脱气;流速为0.3 ml/min;检测波长为304 nm;柱温为35℃,进样量5μl。结果:此方法最低检测浓度为0.018 mg/L,线性范围为0.018~28.200 mg/L,方法平均回收率在95.52%~104.63%,日内及日间精密度的相对标准差均〈10%。结论:该方法简便、快速、重现性好,可用于临床血药浓度监测和复方制剂的药代动力学和生物利用度研究。

Abstract:Objective:To determine salicylic acid in the human plasma by ultra performance liquid chromatography(UPLC).Methods:Salicylic acid was successfully separated using a Waters Spursil C18 column.The mobile phase consisted of acetonitrile-0.2%phosphoric acid(20.7∶79.3) and was degassed ultrasonically before use.Each component of the mobile phase was filtered through a 0.22 μm membrane.The column temperature was 35℃,the flow rate was 0.3 ml/min and detection wavelength was 304 nm.Injection volume was 5 μl.Results:The detection limit was 0.018 mg/L.The linear range was 0.018~28.200 mg/L.The average recovery was 95.52%~104.63%.RSD of intra-day and inter-day was less than 10%.Conclusions:The method is convenient,fast,reproducible.and can be used for determination simultaneously of concentration of salicylic acid in human plasma.It is suitable for therapeutic monitoring and the pharmacokinetics study.
