摘    要:子宫内膜异位症(endometriosis,EMs)是一种病因未明,处理棘手,严重危害妇女健康的疾病。异位内膜的种植和增殖必需有新生血管的形成。在血管形成中血管内皮生长因子(VEGF)起关键作用,因此VEGF与EMs的关系受到重视。目前VEGF基因启动子区多态性与EMs发病关系及采用抗VEGF治疗EMs是研究的热点。5′未翻译区和3′未翻译区均有多个多态性位点,其中以-2578~C,+405G/C,-460T/C和+936C/T最具代表性,其对VEGF基因启动子的活性有很大影响,并可能改变VEGF的量。很多实验证明,针对VEGF及其受体的药物,如抗VEGF抗体等能有效抑制EMs发展。为EMs诊断和治疗提供新的思路。

The Role of Vascular Endothelial Growth Factor in Pathogenesis and Treatment of Endometriosis

Abstract:Endometriosis is a significant debilitating gynaecological disorder but the precise aetiology of that is still not clear. Angiogenesis is a major prerequisite for the initiation and progression of endometriosis. Nowadays, many researchers focus on the association between vascular endothelial growth factor (VEGF), the most powerful angiogenic factor, and end0metriosis. A large number of studies have begun to investigate the potential role of VEGF gene promoter region polymorphism in the development of endometriosis. Single nucleotide polymorphisms (SNPs) in the VEGF 5'untranslated region (5'UTR) or 3'untranslated region (3'UTR) have been reported to be associated with endometriosig in different populations. The representative of functional VEGF polymorphisms, -2578A/C, +405G/C, -460T/C and +936C/T, have an effect upon VEGF gene promoter activity and result in an increase of VEGF levels. And several experiments show the management of antiangiogenic agents, such as antihuman vascular endothelial growth factor (anti-hVEGF), can effectively inhibit the development of endometriosis. These will shed promising light on the diagnosis and therapy for endometriosis.
