摘    要:
PARP-1/AIF通路介导的非caspase依赖性细胞凋亡多见于缺血再灌注或某些药物引起的神经细胞死亡。PARP-1定位于细胞核,参与细胞内多种生理活动。广泛的DNA损伤引起PARP-1过度激活,进而使线粒体蛋白AIF转位至细胞核,作为DNA内切酶引发染色质凝集、DNA片段化和细胞死亡。最近,对该通路信号转导和调控机制的研究取得了快速的进展。

Abstract:
PARP-1/AIF-mediated caspase-independent apoptosis plays an important role in ischemia/reperfusion or drug-induced death of neural cells. PARP-1 is located in nucleus and participates in various physiological processes. Extensive DNA damage causes overactivation of PARP-1 which induces, in turn, the translocation of AIF from mitochondria to nucleus where AIF functions as a DNA endonuclease, leading to chromatin condensation, DNA fragmentation and apoptosis. Recently, rapid progress has been made in the study of the molecular mechanism of signal transduction and regulation.
