辛伐他汀对PC12细胞糖氧剥离损伤的保护作用及其机制探讨

摘    要:目的探讨不同浓度的辛伐他汀干预对PC12细胞糖氧剥离损伤的保护作用及其作用机制。方法对PC12细胞进行1h的缺糖缺氧损伤和24h的再灌注操作,并在损伤前1h加入不同浓度的辛伐他汀进行干预,并观察PC12细胞的形态和结构,MTT测细胞活力,检测一氧化氮(NO)、白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)的浓度。结果PC12细胞在1h的缺糖缺氧损伤和24h的再灌注后与对照组比较,其细胞的突起减少或消失,细胞肿胀变圆,聚集成团;给予辛伐他汀干预的PC12细胞能够明显对抗糖氧剥离和再灌注的影响;对照组和糖氧剥离+不同浓度的辛伐他汀组的细胞活性均明显高于糖氧剥离组(P〈0.01);对照组和糖氧剥离+不同浓度的辛伐他汀NO、IL-1β和TNFua的浓度均低于糖氧剥离组(P〈0.01)。结论通过对PC12细胞进行1h的缺糖缺氧损伤和24h的再灌注操作,能够较好地模拟神经细胞在缺血和再灌注后的状态;通过加入不同浓度的辛伐他汀进行干预,能够明显保护PC12细胞糖氧剥离和再灌注的损伤。

Protective effect of simvastatin on PC12 cell in oxygen and glucose deprivation injury

Abstract:Objective To investigate the protective effect of simvastatin with different concentrations on PC12 cell in oxygen and glucose deprivation injury. Methods PC12 cells were treated under oxygen and glucose deprivation(OGD) for 1 hour and reperfusion for 24 hours, and divided into the normal control group, OGD group and OGD with simvastatin groups in which the cells were pretreated 1 hour before OGD by differ ent concentrations. Concentrations of NO, IL-1β and TNF-α in all groups were measured after the reperfusion model of PC12 cell was established, The cell viability was assayed by MTT. Results Concentrations of NO, IL-1β and TNF-α of OGD group significantly higher than those of the other group(P〈0. 01),The cell viability significantly milder in OGD group than those in the other groups (P〈0. 01 ). Conclusions Simvastatin pre treatment with different concentrations has protective effect on PC12 cell in OGD in vitro.
